摘要
目的:观察培养生长于羊膜的角膜缘干细胞移植治疗完全性角膜缘缺陷的效果。方法:建立兔完全性角膜缘缺陷模型,7d后分别给予羊膜移植、自体角膜缘移植、体外培养的角膜缘干细胞移植,比较观察3组治疗后的临床表现。结果:羊膜移植组有明显的新生血管和上皮缺损,角膜浑浊。而自体角膜缘移植和体外培养的角膜缘干细胞羊膜移植组术后角膜上皮光滑完整,无上皮缺损和新生血管新成。结论:自体角膜缘干细胞体外培养羊膜移植是治疗完全性角膜缘缺陷的有效方法,可恢复角膜缘屏障功能。
AIM:To investigate the treatment of rabbit total limbal deficiency by cultured limbal stem cells growing on amniotic membrane for autograft transplantation.METHODS:Right eye of 30 rabbits were rendered totally limbal stem cell deficiency by surgical removal of 360 degree of limbal rim, after 7 days different treatment were performed in rabbits.Group 1(n=10) received amniotic membrane transplantation,whereas group 2(n=10) received limbal autograft transplantation and group 3 underwent autograft transplantation limbal stem cell cultured on human amniotic membrane derived from a small limbal biopsy specimen from the left eye.The corneal changes were observed by a slitlamp.RESULTS:Group 1 rabbits showed a significant corneal neovascularization and epithelial incomplete, but group 2 and group 3 showed a smooth and integrated corneal epithelium and no corneal neovascularization.CONCLUSION:The autograft transplantation of limbal stem cells cultured on human amniotic membrane is an effective method to treat total limbal stem cell deficiency and recover limbal cellular barrier function and reconstruct the ocular surface.
