摘要
目的:探讨碱性成纤维细胞生长因子(bFGF)与骨形态发生蛋白(BMP)复合对节段性骨缺损的修复作用,以及在骨修复过程中bFGF和BMP之间的相互作用。方法:用羟基磷灰石(HA)作为载体,将一定量的BMP和/或bFGF与之复合,形成HA/BMP/bFGF、HA/BMP复合物,分别修复1.0cm兔桡骨节段性缺损。术后2、4、8、12周取材,进行放射学、组织学及新骨形成的定量分析。结果:新骨能长入植入材料孔道内和覆盖植入材料,修复骨缺损。第2、4、8、12周时,植入HA/BMP/bFGF复合材料侧新骨形成量均多于H-BMP侧。结论:BMP/bFGF复合后骨修复作用优于单一的BMP,在骨缺损修复过程中BMP和bFGF有良好的协同作用。
Objective: To evaluate the effect of basic fibroblast growth factor and bone morphgenetic protein composite on the repairing of Segmental bone defects and to discuss the interaction between bFGF and BMP during bone repair. Methods: Segmental bone defects for 1 .0cm were created in middle part of the radial shafts of 28 adult rabbits. The defects were filled with implants of HA/BMP/bFGF, HA/BMP Separately. The defects were examined radiographically and histologically as well as quantitative analysis of the new bone formation at 2、4、 8、 12 weeks post-operation. Results: The new bone of host bed could grow into the pores of implanted material and cover the surfaces . In the ways, the bone defects were repaired and on the side which HA/BMP/bFGF composite was implanted into, new bone formation was rather richer than that with HA/BMP at 2、 4、 8 and 12weeks. Conclusion: These data demonstrated not only the synergistic action between BMP/bFGF in the process of bone healing , but also a better effect of BMP/bFGF composite than single BMP on bone repair.
