摘要
为探讨哮喘大鼠HPA(下丘脑 腺垂体 肾上腺 )轴变化及不同补肾法 (阴阳双补、补肾阴、补肾阳 )对该轴的影响。SD大鼠腹腔注射卵蛋白制成哮喘模型 ,分别予相应方法治疗 ,并与必可酮气雾剂比较。采用逆转录聚合酶链反应和图象分析法、放射免疫法 ,分别观察各组大鼠下丘脑CRHmRNA ,血浆ACTH、CORT水平的变化。结果 :模型大鼠下丘脑CRHmRNA ,血浆ACTH、CORT水平均明显低于对照组 ,补肾三方均能不同程度提高哮喘大鼠下丘脑CRHmRNA表达 ,升高血浆ACTH、CORT水平 ,阴阳双补法作用最为显著 ,补肾阳法作用其次 ,补肾阴法作用不甚明显。结论 :哮喘模型大鼠存在着下丘脑水平的HPA轴的抑制 。
To observe the changes of hypothalamic-pituitary-adrenal (HPA) axis and the effects of different kidney-nourishing methods(nourishing kidney yin and yang,nourishing kidney yin,and nourishing kidney yang) on it.SD rats were modeled into asthmatic rats by abdominal injection of egg albumin,and then treated by aforementioned methods;moreover,Beclomethasone aerosol was used as control.The levels of hypothalamic CRH mRNA,plasma ACTH and CORT were measured by reverse-transcription polymerase chain reaction,graphics analysis and radioimmunoassay.Results:In the asthmatic rats,the levels of hypothalamic CRH mRNA,plasma ACTH and CORT fell obviously,while in the three herbal treatment groups,they increased to different extent,especially in the groups treated by nourishing kidney yin and yang,and nourishing kidney yang.Conclusion:In the asthmatic rats,the hypothalamic HPA axis is inhibited,but nourishing kidney yin-yang or kidney yang can release the inhibition.
