本文以5-甲基-2-甲氧其苯胺(克立西汀)为原料通过N-酰化反应(温度:70℃~80℃,收率:96.2%,纯度:99.6%)、氯磺化反应(温度:0℃-60℃,收率:89.2%,纯度:97.5%)、磺酰胺化反应(温度:70℃-80℃,收率:96.2%,纯度:99.5%)、酰胺水解反应(3mol/l盐酸回流,收率:81.7%,纯度:99.2%)合成标题化合物,讨论了反应条件。
N-methyl-2-methyI-5-methoxy-4-aminobenzene sulfonamide was prepared starting with 5-methyl-2-methoxyaniline (cresidine) by N-acylation (at 70-80℃, yield 96.2%, purity 99.6%), chiorosulfonation (at 0-60℃, yield 89.2%, purity 97.5%), sulfonamidation(at 70-80℃, yield 96.2%, purity 99.5%), and hydrolysis of amides ( refluxing in 3moI/L HCl, yield 81.7%, purity 99.2%). The reaction conditions were discussed.
