摘要
目的 癌胚抗原(CEA)是1种国际上公认的肿瘤标志物,是某些肿瘤诊断和治疗的靶分子,为了增强表达CEA的重组质粒对肿瘤的免疫防治效果,我们构建了1套表达CEA的真核表达质粒。方法运用基因工程技术将编码人CEA全长2.4kb cDNA片段插入到表达载体pCI—neo中,用限制性内切酶法对其进行鉴定。结果 构建的重组质粒含有2.4kb CEA cDNA片段,且正向插入。结论这为更好地研究CEA的重组质粒对CEA阳性肿瘤的免疫效应打下基础。
Objective Carcinoembryonic antigen(CEA) is an important marker of tumor and an effective target molecule for the diagnosis find therapy of some malignant tumors. In order to enhance the anti-tumor effects of recombinant plasmid expressing CEA, eukaryotic expression plasmid encoding CEA cDNA was constructed. Methods Genetic engineering techniques were applied to insert a 2. 4kb fragment containing full-length human CEA into a eukaryotic expression vector pCI-neo.The resultant recombinant plasmid was cleaved with appropriate endonucleases. Results The insertion 2. 4kb fragment was certified to be in the right direction. Conclusions The results pave the way for further research on the immunological therapy of some tumors expressing CEA.
