系统性红斑狼疮患者外周血CD4^+ CD8^+和CD22^+淋巴细胞活化分子CD69的表达 被引量:7

摘要 目的 探讨系统性红斑狼疮 (SLE)患者外周血淋巴细胞 (PBL)T细胞 (CD4 + 、CD8+ )和B细胞 (CD2 2 + )活化分子CD6 9的表达。方法 应用双染色流式细胞术检测CD4、CD8、和CD2 2细胞亚群CD6 9分子 ;在植物凝集素 (PHA)刺激后 2 0h淋巴细胞亚群CD6 9分子的表达。结果 ①SLE患者PBMC的CD6 9分子活动期高于静止期 (P <0 0 0 1)和正常对照组 (P <0 0 1)的表达 ,SLE静止期患者与正常对照组CD6 9表达差异无显著性 (P >0 0 5 )。②进一步分析CD4 + 、CD8+ 和CD2 2 + 淋巴细胞亚群的CD6 9的表达 ,其中 ,SLE活动期患者CD4 + 细胞的CD6 9表达显著高于静止期 (P <0 0 0 1)和正常对照组 (P <0 0 1)的表达 ,SLE静止期患者与正常对照组CD6 9表达差异无显著性 (P >0 0 5 ) ;CD8+ 细胞活动期高于静止期患者 (P <0 0 5 ) ,其余组间差异无显著性 (P >0 0 5 ) ;CD2 2 + B细胞各组间差异无显著性。③PHA刺激 2 0h后 ,CD4 + 、CD2 2 + B细胞的CD6 9表达 ,活动期显著高于静止期患者和正常对照组 (P <0 0 1)。结论 SLE患者外周血CD4 + T细胞和CD2 2 + B细胞存在着异常的活化 ,这种淋巴细胞的异常活化是SLE重要的发病机制之一。 Objective To study the expression of CD69 on CD4 + T cells.CD8 + T cells and CD22 + B cells in peripheral blood lymphocytes (PBL) in patients with systemic lupus erythematosus (SLE).Methods The CD69 molecule was measured on CD4 + T cells,CD8 + T cells and CD22 + B cells in PBL with dual color fluorescence cytometry before and after stimulated by PHA.Results ① The expression of CD69 molecule in PBL of active patients was higher than that of inactive patients ( P <0 01) and that of control subjects ( P <0 01).The expression of CD69 molecule in PBL of inactive patients was not significantly different from that of normal groups.②It was shown that the expression of CD69 molecule on CD4 + T cells in PBL of active patients was higher than that of inactive patients ( P <0 01) and that of control subjects ( P <0 01) in further study.The expression of CD69 molecule on CD8 + T cells in PBL of active patients was higher than that of inactive patients ( P <0 05).The expression of CD69 molecule on CD22 + B cells in PBL was not significantly different among active,inactive patients and control group ( P >0 05).③The expression of CD69 molecule on CD22 + B cells and CD4 + T cells in PBL of active patients was significantly different from inactive patients and control group ( P <0 01) after 20 hours stimulated by PHA.Conclusion The increase of CD69 molecule on CD4 + T cells and CD22 + B cells in the PBMC of SLE is certainly associated with pathogenesis of SLE.
