摘要
目的 :探索简便、经济的大鼠酒精性肝纤维化造模方法。方法 :3 3只大鼠随机分为 2组。模型组予酒精灌胃 ,于实验的第 4、12、2 4周末分批采血 ,以测定生化指标 ,后行放血法处死。将取出的肝脏进行常规切片、HE染色及Masson三色胶原染色后行病理学观察。正常对照组以等量饮用水代替酒精灌胃 ,于实验第 2 4周末处死 ,处理同模型组。结果 :于实验第 2 4周时观察到了除酒精性肝硬化外酒精性肝病的各种病理表现 ,并且随着酒精刺激时间的延长肝纤维化程度逐渐加重。结论 :应用酒精灌胃法可成功复制大鼠酒精性肝纤维化模型 ,该方法简便。
Objective To induct a relatively simple and feasible rat model for further pathogenesis of alcoholic liver fibrosis.Methods Thirty-three Sprague-Dawley rats were categorized into two groups.Twenty-seven rats were intragastrically infused with ethanol while six rats in control group were infused with the same quantity of water.Nine rats respectively in model groups were randomly sacrificed at the end of 4th,12th and 24th week after experiment,blood samples were collected to measure the concentrations of blood proteins and enzymes.Hematoxylin-eosin(HE)method was used to examine the degree of hepatic steatosis and the alcoholic hepatitis.Masson's trichrome(MT)method was used to observe fibrous tissue.Results Many kinds of pathologic changes except alcoholic cirrhosis can be observed in alcoholic liver disease at the end of 24th week after experiment.Along with time passing by,liver fibrosis becomes severe gradually.Conclusion A rat model of alcoholic liver fibrosis may be sufficiently induced by repetitive administration of alcohol.It is simple and feasible.
