摘要
目的:在实验性大鼠酒精诱导肝损伤中研究肝细胞凋亡与Fas、Fas配体(FasL)表达的关系。方法:改进的酒精灌胃法建立大鼠肝损伤模型,应用末端转移酶标记技术(TUNEL)检测肝细胞原位凋亡的情况,免疫组化法分别检测肝组织中Fas、FasL。结果:肝损伤模型组及正常对照组肝细胞凋亡指数分别为(2.03±1.61)%及(0.10±0.21)%(P<0.05)、Fas阳性细胞数分别为7.23±1.10及0.58±0.17(P<0.05)、FasL阳性细胞数分别为5.14±0.76及0.31±0.25(P<0.05)。结论:酒精可以诱发大鼠肝细胞凋亡,肝细胞凋亡的发生与Fas的表达增加有关。
Objective:To study the relationship between hepatocyte apoptosis and expression of Fas and FasL in alcohol- induced liver injury in rats and their effects on the pathogenesis of alcoholic liver disease (ALD). Methods:The alcohol - induced liver injury model was established in rats. The TUNEL assay was used to examine the hepatocyte apoptosis index, and the Fas & FasL expressions were detected by immunohistochemistry. Results: The apoptosis indexes in pathologic model group and control group were (2.03 ± 1.61 )% and (0.10 ± 0. 21 )% respectively ( P 〈 0. 05). The cell numbers of the positive Fas and FasL expressions in pathologic model group and control group were 7.23 ±1.10 vs0. 58 ±0.17 (P〈0.05) and 5.14 ±0.76 vs0. 31 ±0.25 (P〈0.05), respectively. Conclusion: Chronic and excessive alcohol consumption may cause liver injury in rats, which may induce hepatocyte apoptosis. The hepatocyte apoptosis may be related with the up - regulation of the Fas & FasL expressions.
