盐胁迫短芒大麦甜菜碱醛脱氢酶基因片段的克隆与序列分析 被引量:2

摘要 以 2 %NaCl诱导 3~ 6天的短芒大麦叶片为材料 ,用TRIzol试剂提取总RNA后 ,反转录合成cDNA ,用PCR扩增得到约 6 0 0bp片段 ,将所得序列与GenBank上已登录的序列进行同源性比较 ,发现它与大麦等作物的BADH基因同源性很高 ,并存在醛脱氢酶的保守十肽区 ,从而证明短芒大麦的耐盐性与小分子渗透物质的积累有关 . Total RNA was isolated with TRIzol reagent from leaves of Hordeum brevisubulatum ,which was treated with 2% NaCl for 3~6 days.After cDNA synthesized by reverse transcription,a fragment of 600bp was obtained through PCR.Comparison of the nucleotide was performed with those reported in Genbank,The results showed that it has high homology of the nucleotide with that of Barley,and there was a ten peptides conservative region of aldehyde dehydrogenase in this sequence.All the results suggested that the highly salt-tolerance of Hordeum brevisubulatum is correlative with osmotic materials.
