血管内皮生长因子及其受体随增龄在大鼠肾组织内表达的研究 被引量:14

Research of the expression of vascular endothelial growth factor and its receptors in renal tissue of rats with aging

摘要 目的 研究血管内皮细胞生长因子 (VEGF)及其受体Flt 1和Flk 1在大鼠肾组织内的表达及随增龄变化 ,探讨它们在肾脏衰老过程中的作用。 方法 应用 3、12、2 4月龄 (各 7只 )大鼠肾组织石蜡切片进行常规病理及免疫组织化学染色 ,定量分析肾组织内微血管变化及VEGF、Flt 1和Flk 1表达变化。应用逆转录聚合酶链反应技术 (RT PCR)检测肾组织内VEGF AmRNA的表达。 结果  2 4月龄组与 3月龄组相比肾小球面积增大〔(15 6 35± 10 2 2 ) μm2 vs(72 0 5± 496 ) μm2 ,P <0 0 1〕 ,肾小球内毛细血管袢腔面积与肾小球面积百分比减少 (46 76 %± 4 91%vs 6 3 75 %±6 0 2 % ,P <0 0 1) ,肾小管周围毛细血管数量减少 (9 8± 2 6vs 14 7± 3 1,P <0 0 1) ;肾小球内VEGF阳性细胞数增多 (9 3± 2 4vs 6 4± 1 6 ,P <0 0 5 ) ;集合管中VEGF的表达则明显减少(9 35 %± 2 10 %vs 15 2 3%± 3 2 2 % ,P <0 0 5 ) ;Flk 1在肾小球血管袢上表达增加 (9 17%±2 0 2 %vs 1 0 3%± 0 35 % ,P <0 0 1) ,而Flt 1和Flk 1在肾小管上表达则明显减少 (7 6 4%±3 0 2 %vs 15 36 %± 2 5 4% ,2 48%± 0 86 %vs 9 0 1%± 2 6 3% ,P <0 0 1)。 2 4月龄组VEGF AmRNA较其他两组减少 (P <0 0 5 )。 结论 VEGF。 Objective To investigate the role of vascular endothelial growth factor(VEGF) and its receptors Flt-1 and Flk-1 in the aging process of kidney ,we studied the expression of them in renal tissue of rats with aging. Methods Paraffin sections of renal tissue of rats at the age of 3 months old (n=7) ,12 months old(n=7) and 24 months old (n=7)were stained by routine methods and immunohistochemistry. Microvascular changes and the expressions of VEGF,Flt-1 and Flk-1 were quantificationally analyzed. VEGF-A mRNA in kidney was detected by RT-PCR. Results Compared 24 month old group with 3 month old group,it was found that the areas of glomerular were increased〔(15 635±1 022)μm2 vs ( 7 205±496)μm2,P<0.01〕,the percentage of the area of capillary lumen compared with that of glomerulus was decresed(46.76%±4.91% vs 63.75%±6.02%,P<0.01),the number of peritubular capillary was decreased(9.8±2.6 vs 14.7±3.1,P<0.01),the number of VEGF positive cells in glomerular was increased(9.3±2.4 vs 6.4±1.6,P<0.05),but the expression of VEGF in collecting duct cells was decreased(9.35%±2.10% vs 15.23%±3.22%,P<0.05),Flk-1 was expressed increasingly in capillary loops of glomerular(9.17%±2.02% vs 1.03%±0.35%,P<0.01),but the expressions of Flt-1 and Flk-1 in tubular cells was significantly decreased(7.64%±3.02% vs 15.36%±2.54%,2.48%±0.86% vs 9.01%±2.63%,P<0.01).Expression of VEGF-A mRNA in 24 months old group was decreased(P<0.01). Conclusions The expressions of VEGF and Flk-1 were increased in glomerular of rats,but they were decreased in renal tubules of rats with aging,which suggest that the abnormal changes of VEGF and its receptors may play an important role in the aging process of kidney.
