摘要
采用逆转录聚合酶链反应(PCR)技术自SD大鼠肝细胞中克隆出全长1125个bP的编码主要组织相容性抗原Ⅰ类复合体分子(MHC-Ⅰ)cDNA。经限制性内切酶图谱分析证实后,定向插人表达载体pBV220,并筛选出带有插人片断的阳性克隆。为研究在原核或真核表达系统中的表达,及其在抗原识别,免疫应答中的作用奠定了基础。
The total RNA was extincted from the rat liver cells. Accolading to the puLblished seqUence of rat MHC- I(rMHC- I ), a couple of primers for rMHC- 1 was designed and synthesized. POlylnerase chain reaction for reversetranSCription obtained a specific band of rMHC- 1 cDNA fragment from rat liver cells. The obtained fragment was inserted to PBV220 plasthed and tranSformed to E. colt DH5a. Restrichon e^e digestion and PCR identified it as rat rMHC1 with the whole ~ng flame of 1 125 base pairs.
