同型半胱氨酸对人胚心肌细胞HCY-2表达的影响 被引量:4

EFFECT OF HOMOCYSTEINE ON HCY-2 EXPRESSION OF HUMAN EMBRYONIC VENTRICULAR MYOCYTES

摘要 目的 研究同型半胱氨酸对人胚心肌细胞同型半胱氨酸诱导基因 (HCY 2 )表达及对心肌细胞增殖的影响。 方法 取人胚心肌细胞培养 ,采用免疫组织化学及图像分析技术对心肌细胞内HCY 2表达及心肌细胞增殖的变化进行定量测定分析。 结果 当同型半胱酸的浓度小于 1 2 5mmol L时 ,HCY 2的表达随着同型半胱氨酸浓度的增高而逐渐增强 ,并且同型半胱氨酸促进细胞的增殖 ;当浓度超过 1 2 5mmol L时 ,HCY 2的表达逐渐降低 ,同时同型半胱氨酸抑制细胞的增殖。 结论 HCY 2高表达时同型半胱氨酸对心肌细胞的增殖起促进作用 。 Objective To study the effect of homocysteine on homocysteine induced gene(HCY 2) expression and proliferation of human embryonic ventricular myocytes. Methods Human embryonic ventricular myocytes culture,immunohistochemistry and the technique of pictures assay were used to study the effect of homocysteine of HCY 2 expression and analyze the proliferation of human embryo ventricular myocytes quantificationally. Results When the concentration of homocysteine was less than 1 25?mmol/L, the expression of HCY 2 and the cell proliferation increased as the concentration of homocysteine increased; it inhibited the cell proliferation and the expression of HCY 2 when the homocysteine exceeds 1 25?mmol/L.Conclusion The cell proliferation was stimulated by the high expression of HCY 2 and was inhibited by the low expression of HCY 2.
