微波修复CD_(44)s、CD_(44v6)抗原及其在颅内转移瘤与胶质母细胞瘤中表达研究 被引量:1

Study of the Antigen Retrieval By Microwave and Expression of CD_(44)s and CD_(44v6) Genes Protein in Human Intracranial Metastastic Tumors and Glioblastomas

摘要 目的 探讨微波抗原修复对CD44s和CD44v6抗原的影响及其在颅内转移瘤及胶质母细胞瘤中的表达与这些肿瘤侵袭、转移之间的关系。方法 应用微波 -0 1%TritonX -10 0 (MW -T)和微波 -柠檬酸缓冲液 (MW -CA)修复抗原 ,微波 -LSAB免疫组化染色检测CD44s和CD44v6在 2 0例正常脑组织、3 5例脑胶质母细胞瘤和 3 0例颅内转移瘤中CD44的表达情况。结果 MW -CA和MW -T抗原修复处理的IOD显著高于未修复组的IOD值 (P <0 0 1) ;2 0例正常脑组织中CD44s和CD44v6表达均为阴性 ;3 5例脑胶质母细胞瘤CD44s阳性表达率为 10 0 % (3 5 /3 5 ) ,CD44v6表达为阴性 ;3 0例颅内转移瘤中CD44s阳性表达率为 86 7% (2 6/3 0 ) ,CD44v6阳性表达率为 66 7% (2 0 /3 0 )。CD44v6在颅内转移瘤与脑胶质母细胞瘤的表达差异有显著性 (P <0 0 1)。结论 微波抗原修复处理可以显著提高CD44s和CD44v6的IOD值 ;脑胶质母细胞瘤中CD44V6的表达可能与其脑内侵袭过程有关 ,无CD44v6表达可能与其很少发生颅外转移有关 ,并有可能成为颅内转移瘤诊治的有用指标之一。 Objective To investigate the effect of antigen retrieval by microwave and the expression of CD 44 s and CD 44v6 in human intracranial metastastic tumor and human glioblastomas, and the relationship between the expression of CD 44 adhesion molecules and the invasion or metastasis of these tumors.Methods After antigen retrieval by microwave-0.1%Triton X-100(MW-T) and buffer solution of microwave-citrate salts(MW-CA), CD 44 s and CD 44v6 were used to detect the expression of CD 44 adhesion molecules in 20 normal human brain tissues, 35 human glioblastomas, and 30 intracranial metastastic tumor by microwave-LSAB immunohistochemical staining technique.Results Integral optical density(IOD) value of CD 44 s and CD 44v6 positive cells restored by MW-T and MW-CA was higher significantly than that of without antigen retrieval (P<0 01). Expression of CD 44 s and CD 44v6 in 20 normal brain tissues were negative.All 35 glioblastomas(100%) and 66 7%(26/30) in intracranial metastastic tumors expressed CD 44 s.No glioblastomas and 66.7%(20/30) expressed CD 44v6 , showed a significant difference in CD 44v6 expression between glioblastomas and intracranial metastastic tumors(P<0 01). Conclusions IOD value of CD 44 s and CD 44v6 positive cells could be improved significantly by MW-T and MW-CA. CD 44 s may play an important role in the invasion of glioblastomas,and CD 44v6 expression in human glioblastomas may be related to metastases intracranially.CD 44v6 expression may be a useful marker for diagnosis and treatment of patients with brain metastases.
