摘要
用ZymosanA对中国对虾(Penaeuschinensis)血细胞进行体外刺激,用NBT还原法测定血细胞吞噬活动中产生O-2的产生;不同浓度2的过程。结果表明,中国对虾血细胞在经刺激诱导的吞噬活动中有明显的呼吸爆发和O-的ZymosanA对血细胞产生O-2的影响不同。ZymosanA在0.25~1.0mg/mL时随质量浓度的增加,产生O-2的量也增加,但当ZymosanA质量浓度继续增加到1.5和2.0mg/mL时,产生O-2的量反而下降。用同一浓度的ZymosanA对不同密度血细胞进行刺激产生O-2的强度也不同,随着密度的增加,血细胞产生O-2的量增加。SOD、NEM和碘代乙酰胺(Iodoacetamide)对血细胞吞噬过程中O-2的产生有抑制作用;Hg2+、Cr3+、Pb2+、Cu2+和Zn2+5种重金属离子也能抑制血细胞产生O-2;不同个体的中国对虾血细胞产生O-2的能力不同。NBT还原法测定中国对虾血细胞吞噬时产生的O-2可以作为检测对虾免疫状态的指标。
The hemocytes of Penaeus chinensis were stimulated in vitro by Zymosan A and the production of superoxide (O2) was detected by nitroblue tetrazolium (NBT) reduction assay. The results indicate that the hemocytes of P. chinensis can have an obvious burst of respiratory and generation of O2during the phagocytic activity after the stimulation. Zymosan A at different concentration has different effects on the O2 production that when Zymosan A is at 0.25-1.0 mg/mL, the O2 production increases with the increase of Zymosan A concentration, and when Zymosan A at 1.52.0 mg/mL, the O2 production decreases. When Zymosan A is at the same concentration, the stimulated hemocytes at different cell dense can generate different amount of O2. SOD, NEM and Iodoacetamide can inhibit the hemocyte activity of NBT reduction during the phagocytosis of hemocytes, and the other five metal ions like Hg2+, Cr3+, Pb2+, Cu2+ and Zn2+ also have the same function. The abilities of O2 generation among the individuals of P. chinensis are highly different. It suggests that the hemocytes of P. chinensis could generate reactive oxygen species like O2 to kill their pathogens in the phagocytec defensive reaction.
