铅对背角无齿蚌抗超氧阴离子与抑制羟自由基的影响以及可溶性蛋白分析 被引量:5

摘要 采用浓度为0.2,0.3,0.5 mg.L-1的铅溶液对背角无齿蚌(Anodonta woodiana)进行处理,研究铅离子对蚌的抗超氧阴离子(O2-)、抑制羟自由基(OH-)以及可溶性蛋白的影响。结果发现,随着铅离子的浓度升高,蚌血清的抗超氧阴离子及抑制羟自由基活性均有明显升高。肝胰腺中抗超氧阴离子活性随着铅离子浓度的增高呈现先降后升的趋势。肝胰腺中抑制羟自由基的活性也是先降后升。血清及肝胰腺中可溶性蛋白含量随着铅离子浓度的增高均呈现先升后降的趋势。鳃中可溶性蛋白含量随铅离子浓度的增高也呈先升后降的趋势。用铅溶液处理蚌后,对蚌的血清、肝胰腺和鳃进行SDS-PAGE电泳发现,血清、肝胰腺和鳃中均有正常谱带的缺失;表明外界环境中的铅离子可能严重干扰机体内蛋白质的生物合成,引起了蛋白变性,错误折叠和蛋白聚合。 This paper studied the activity of resisting superoxide anion (O2^-) and scavenging hydroxyl free radical ( OH^- ) of Anodonta woodiana induced by Pb^2+ of concentration 0.2,0.3,0.5 mg · L^-1. The results showed that the activities of both resisting superoxide anion ( O2^- ) and scavenging hydroxyl free radical ( OH^- ) from Anodonta woodiana hemolymph were raised significantly with the increased Pb^2+ concentration. The activity of resisting superoxide anion ( O2^- ) of hepatopancreas was decreased first and then increased with the raised Pb^2+ concentration. The activity of resisting superoxide anion ( O2^- ) of hepatopancreas were both higher than the control group at the other concentration. The activity of scavenging hydroxyl free radical ( OH^- ) of hepatopancreas was also decreased first and then increased with t the raised Pb^2+ concentration. The soluble protein content of hemolymph and hepatopancreas were both increased first and then decreased; the soluble protein content of gills was also increased first and decreased afterwards with the raised Pb^2+ concentration. SDS - PAGE of the hemolymph, hepatopancreas and gills from Anodonta woodiana were different. It indicated that Pb^2+ might interfere protein biosysnthesis in Anodonta woodiana.
