摘要
目的 探讨全耳蜗灌流钙通道阻滞药对耳蜗微音电位 (CM)输入 输出函数曲线的影响。方法 应用豚鼠全耳蜗灌流技术 ,耳蜗灌流不同浓度的钙通道阻滞药尼福地平 ,观察记录各声压级CM幅度并绘制输入 输出函数 (input output,I O)曲线。结果 耳蜗灌流人工外淋巴液CM相对幅度和I O函数曲线非线性无改变 ;耳蜗灌流0 .15 μM尼福地平后 ,CM相对幅度轻度下降 ,但I O曲线非线性无变化 ;耳蜗灌流 0 .5 μM、3 μM尼福地平后CM高声压级的相对幅度明显降低 ,I O函数曲线非线性特点减弱。结论 钙通道阻滞药可通过作用于外毛细胞上的钙通道影响耳蜗功能 ,且与剂量相关 ;同时间接证明外毛细胞上存在L
Objective To investigate the influence of calcium channel blocker(nifedipine)on the input/output(I/O)functions of cochlear microphonics(CM).Methods The cochlear microphonics were recorded from round window(RW)before and after the animals were perfused in whole cochlear with different concentration of nifedipine and the input/output functions of CM were analysed.Results The artificial perilymph did not induce a change of the amplitude of CM and I/O function.After the perfusion of 0.15 μM nifedipine,the amplitude of CM reduced slightly,but I/O function did not change.After the perfusion with 0.5 μM and 3 μM nifedipine,the amplitude of CM reduced obviously and nonlinearly.I/O functions also decreased.Conclusion The cochlear microphonics(CM) could be affected by calcium channel blocker with blocking the calcium channel on the outer hair cells (OHC),and the effect correlated to the dose of nifedipine.It is indirectly proven that there is L-calcium channel on the OHC.
