摘要
目的采取人工外淋巴灌流方法建立动物模型,是耳蜗生物学特性研究的新方法,但其成功率较低。为提高人工外淋巴灌流实验的成功率,本实验探索和改进了灌流管的封固和灌流速度等实验技术。方法通过安放圆窗电极监测由短声诱发的听神经复合动作电位(compoundactionpotentialCAP)阈值、扫描电镜观察动物颞骨标本,在生理功能和形态学上予以验证此方法的可靠性。结果灌流前CAP阈值41.00±3.16dBSPL,灌流2小时(或少数动物2.5小时)后为42.00±2.18dBSPL,CAP阈值灌流前、后未见明显变化。扫电镜观察内、外毛细胞的形态结构与对照耳相比未见明显变化。结论生理指标及形态学观察结果均证明,改进后的灌流方法可行。
Objective To improve technique of sealing the perfusion tube and speed of Perfusion, tomake animal model by Perfusion with artificial Perilymphatic fluid. Methods To record compound Action Po-tential (CAP)threshold with click with electrical prober at round window, to observe morphology by Scan Elec-trode Microscopy(SEM) for proving animals' physiological function and morphology. Results Before Perfu-sion CAP's threshold was 41.00±3.16 dB SPL,after 2 h(a few animals 2.5 h)perfusion CAP's threshold was42.00±2. 18 dB SPL. There were not positive changes between before and after Perfusion on function.Cochlear morphology had not been changed after 2 h(a few ammals 2. 5 h) Perfusion to compare with normal leftitself. Conclusion So experiment techniques after improvement are well.
