脐血CD34^+细胞巨核系定向诱导分化的研究

摘要 目的 :研究TPO与SCF、IL - 11协同在体外促进入脐血CD34+ 细胞向巨核系增殖分化的效应。方法 :利用流式细胞仪分离纯化脐血CD34+ 细胞 ,在含血清液体培养体系中、细胞因子诱导下培养 14d。采用流式细胞术测定不同时间点培养体系中CD4 1+ 细胞的比例 ;同时采用甲基纤维素半固体集落培养测定CFU -MK的数量。结果 :经 14d培养 ,TPO +SCF +IL - 11组可使CD4 1+ 细胞和CFU -MK数量分别扩增 4 0 .83± 12 .4 1倍、7.86±2 .6 3倍 ,明显高于TPO组的 2 8.6 9± 8.2 7倍、4 .4 8± 1.2 5倍。结论 :在体外SCF和IL - 11可以协同增强TPO诱导脐血CD34+ Objective:To study the capacity of the combination of TPO+SCF+IL-11 in inducing umbilical cord blood CD34 + progenitor cells to proliferate and differentiate committedly to megakaryocytes.Methods:CD34 + cells were isolated from umbilical cord blood by fluorescence activated cell sorting(FACS) and cultured with cytokines in a serum-containing liquid culture system for 14 days. At certain intervals.CD41 antigen expression and CFU-MK were studied by FACS and megakaryocytic colony forming assays,respectively.Results:In TPO+SCF+IL-11 group,the number of CD41 +cells and CFU-MK were remarkably greater than that in the group of TPO (40.83±12.41 fold vs 28.69±8.27 fold;7.86±2.63 vs 4.48±1.25,respectively).Conclusions:In vitro SCF and IL-11 can synergize with TPO in inducing umbilical cord blood CD34 + cells to proliferate and differentiate committedly to megakaryocytes.
