丽格海棠组织培养外植体再生体系建立关键技术的研究 被引量:21

摘要 以丽格海棠的叶、茎、花茎、叶柄等营养器官作为外植体进行组织培养,着重研究丽格海棠的最佳外植体,最佳培养基配方及接种模式的关键技术。研究表明初代培养基本培养基为MS,激素配比为6-BA0 5mg/L+NAA0 1mg/L,培养30d可获得较多的不定芽。外植体分化能力的研究表明丽格海棠的不同类型外植体能力大小为:叶>茎>花茎>叶柄;从叶片的不同部位来看,叶中>叶侧>叶边缘;从不同叶龄来看,中等成熟叶>幼叶>完全成熟叶>芽叶。叶片的接种模式:反放比正放好。 Leaves, sections of stems, flower stems, leafstalks of Rieger Begonia were used as explants in study of organ culture to establish a rapid technique system of propagation in vitro, and to find out the best explants, medium culture procedure. The results are as follows: Iniation medium: MS+6-BA 05?mg/L+NAA 01?mg/L adventitious buds can be got after 30 days culture. Effects of different explants on shoot regeneration: leaf>stem>flower stem>leafstalk. Effects of different parts leaves on shoot regeneration: middle leaf>lateral of leaf>edge of leaf. Effects of different age of the leaves on shoot regeneration: mediummature leaf>immature leaf>mature leaf>bud leaf. Effect of leaf inoculation model on shoot regeneration: while the leaves are placed on regeneration media with the abaxial surface in contact with the media, more adventitious shoots are obtaind from the leaves; While the leaves are placed on regeneration media with the adaxial surface in contact with the media, less adventitious shoots are obtaind from the leaves.
