兔额部撞击伤后视神经血栓素A_2和前列环素的变化 被引量:1

The content of thromboxane and prostacyclin in optic nerves after forehead impact injury in rabbits

摘要 目的 观察家兔额部撞击伤后视神经血栓素 A2 (thromboxane,TXA2 )和前列环素 (prostacy-clin,PGI2 )的变化。 方法 小型生物撞击机以高压气仓压力 5 113mm Hg(1mm Hg=0 .133k Pa)撞击 32只家兔右侧额骨 ,出现传入性瞳孔障碍的 2 4只兔纳入实验 ,随机分为 1、2、4、7d组。右眼为实验组 ,左眼为对照组。观察撞击伤前后兔眼闪光视觉诱发电位 (flash visual evoked potential,F- VEP)变化 ,并于伤后 1、2、4、7d分别摘除兔眼取视神经做病理检查并检测 TXA2 、PGI2 产物 TXB2 、6 - Keto- PGF1α的含量。 结果 出现传入性瞳孔障碍的 2 4只兔眼经病理检查证实有视神经损伤。其伤后 F- VEP的 P1 波潜伏时与伤前相比显著延长(P<0 .0 1)。 F- VEP的 P1 波振幅显著降低 (P<0 .0 1)。伤后第 1天视神经 TXB2 的含量为 (172 .35± 2 6 .5 2 )pg/ mg、6 - Keto- PGF1α的含量为 (16 1.78± 2 4 .83) pg/ mg,比对照组的 TXB2 含量 [(10 1.6 4± 7.94 ) pg/ mg]和6 - Keto- PGF1α含量 [(0 .85 6± 0 .0 5 ) pg/ mg]显著升高 (P<0 .0 1)。TXB2 与 6 - Keto- PGF1α比值 (1.0 77± 0 .18)与对照组相比亦显著升高 (P<0 .0 5 ) ,直至伤后第 7天仍高于对照组。 结论 额部撞击伤后视神经 TXA2 、PGI2 含量显著提高 。 Objective To observe the content of thromboxane (TXA 2) and prostacyclin (PGI 2) in optic nerves after forehead impact injury. Methods The right forehead zones of 32 rabbits were struck by biology impact machine. Tweenty four rabbits that had afferent papillary defect after injury were chosen, and randomly divided into four groups: 1 day, 2, 4, and 7 days group. Right eyes were in the experimental group and left eyes were in the control group. Flash visual evoked potentials were examined before and after the traumatic injury. The rabbits′ eyes were removed, the optic nerves were pathologically examined, and the content of TXB 2 and 6 Keto PGF 1α which were the products of TXA 2 and PGI 2 were assayed 1, 2, 4, and 7 days after traumatic injury respectively. Results Histopathological examination revealed the findings of injuries of optic nerves of all the 24 rabbits. The latency of wave P 1 was significantly delayed after traumatic injury ( P <0 01), and amplitude of wave P 1 was significantly decreased after traumatic injury ( P <0 01). The content of TXB 2 [(172.35±26.52) pg/mg] and 6 Keto PGF 1α [(161.78±24.83) pg/mg] were significantly higher in the injured optic nerves than in the uninjured ones 1 day after the traumatic injury ( P <0.01). The rate of TXB 2/6 Keto PGF 1α (1.077±0.18) was significantly increased compared to the control group ( P <0.05), and lasted to the 7th day. Conclusions The content of TXA 2 and PGI 2 significantly increases and the ratio of them is lopsided after forehead impact injury in rabbits.
