摘要
目的 探讨胸腺肽α 1 抗肿瘤的机制。方法 用间接酶联免疫吸附试验检测经胸腺肽α1 刺激的8份正常人外周血单个核细胞培养上清液中的白细胞介素2 (IL-2)、肿瘤坏死因子α(TNFα)、可溶性TNFα受体(sTNFαR)和白细胞介素12 (IL-12)含量 ,集落培养检测15份骨髓捐献者骨髓单个核细胞粒单细胞集落形成单位(CFU -GM)、红细胞集落形成单位 (CFU-E)。直接细胞计数法、MTT法检测胸腺肽α 1 对K562细胞的直接抑制作用。结果 胸腺肽α1 能刺激正常人外周血单个核细胞产生IL-2 TNFα、sTNFαR和IL -12增多 ;胸腺肽α1 浓度≥1μg/ml时CFU-GM、CFU -E集落数增多36.9%和70.3 %(P<0.001) ;同时胸腺肽α 1 能直接抑制K562细胞的生长。 结论 胸腺肽α 1 能促进造血干细胞增生 ,通过细胞因子途径和直接抑制作用发挥其抗肿瘤作用。
Objective To explore the antitumor activity of thymosin α1 (Tα1).Methods IL-2,TNFα,sTNFαR and IL-12 levels in the culture supernatants of peripheral blood MNC were measured with indirect ELISA assay from 8 normal volunteers and CFU-GM and CFU-E of bone marrow MNC were detected with colonogenic growth assays from 15 normal volunteers after Tα 1 treatment.Using MTT assay in vitro, to detect the direct inhibition of Tα 1 on K652 cells.Results After Tα 1 treatment the production of IL-2,TNFα,sTNFαR and IL-12 increased;when the concentration of Tα 1≥1μg/ml, the numbers of CFU-GM and CFU-E increased by 36.9% and 70.3% respectively compared with those in controls(P<0.001).Tα 1(1~2μg/ml)inhibited growth of K562 cells significantly in 24 hours and 48 hours.Conclusion The date suggest that Tα1 could promote hemopoiesis and present antitumor activity by enhancing cytokine production and direct inhibition.
