摘要
目的 利用转化生长因子 - β(TGF β)多克隆抗体 (多抗 )阻断TGF β生物学作用 ,观察TGF β多抗预防腹腔粘连的效果。 方法 建立大鼠术后肠粘连模型 ,分别腹腔注射生理盐水 (对照组 )、透明质酸钠 (透明质酸钠组 )和不同剂量的TGF β多抗 (抗TGF β组 ) ,术后 2 1d进行粘连评分 ,其中 30只大鼠分别于术后 3 ,1 0d处死 ,取粘连部位组织 ,用免疫组化法 ,测定TGF β的表达。 结果 术后 2 1d粘连评分 :抗TGF β组为 (2 .4± 0 .99) ,显著低于对照组 (6 .0± 1 .2 5)和透明质酸钠组 (3 .4±1 .0 3) ;在不同TGF β多抗浓度中 50 μg组较经济有效 ;对照组和透明质酸钠组TGF β的表达有时间依从性。术后 3d达高峰 ,这种表达可被TGF β多抗抑制。 结论 TGF β多抗在动物模型上可预防腹腔粘连的发生 ,其机制在于抑制了TGF
Objective To block the bio function of TGF β by the application of anti TGF β polyclonal antibody, and observe its efficacy of prevention of abdominal adhesion. Methods The postoperative abdominal adhesion model was established in SD rats. The drugs were administrated by abdominal injection with saline (control group), sodium hyaluronate( HA group), and varied dosage of anti TGFβ (anti TGFβ groups) respectively. The adhesion was scored 21days later, while 30 of them were executed on the day 3 and day 10 after operation respectively. The expression of TGF β was checked by immunohistochemistry in the samples obtained from the adhesion sites. Results The score of adhesion in anti TGF β group (2.4±0.99) was significantly lower than that in control group (6.0±1.25) and HA group (3.4±1.03); in different dosage of anti TGF, the 50μg group showed its economical efficacy; in the control and HA groups the expression of TGF β had a time dependent manner, which reachs to maximum in the day 3, and could be reduced by antibody. Conclusions The polyclonal antibody of TGF β shows the power to prevent the postoperative abdominal adhesion in animal model, the mechanism of which is due to inhibition of TGF β expression.
