顺铂作用下沙鼠耳蜗细胞Bcl-2及Bax的表达 被引量:1

Expressions of Bcl-2 and Bax protein in Mongolian gerbil cochlear following cisplatin administration

摘要 目的 :通过检测应用顺铂 (CDDP)前后凋亡相关蛋白 Bcl- 2蛋白家族中 Bcl- 2和 Bax在沙鼠耳蜗 Corti氏器和螺旋神经节 (SG)神经元的表达情况 ,探讨 Bcl- 2蛋白家族是否参与了 CDDP诱导耳蜗细胞凋亡。材料和方法 :对沙鼠进行顺铂连续腹腔注射 ,4 mg/ kg,分别于给药 4、5、6、7天组各处死沙鼠 10只 ,取左侧耳蜗 ,做平行蜗轴的石蜡切片 ,用免疫组织化学方法检测连续用药不同时间后底转 SG及 Corti氏器的 Bcl- 2及 Bax的表达变化情况。结果 :在用药 4~ 7天中 ,耳蜗底转 SG及 Corti氏器的 Bcl- 2表达不断下降 ,分别在用药第 6天和第 7天时低于正常 ;Bax在用药第 5~ 7天时显著高于正常。结论 :CDDP作用下 ,Bcl- 2和 Bax表达发生变化 ,Bcl- 2蛋白家族参与了 CDDP诱导的耳蜗细胞凋亡。 Objective: To investigate the expression of Bcl 2 and Bax proteins,regulators of apoptosis,in Mongolian gerbil cochlear injury by cisplatin(CDDP).Methods:The cisplatins were administred 4mg/kg/day on Mongolian gerbil consecutively for 4 to 7 days.The expressions of Bcl 2 and Bax proteins were examined by immunohistochemical method in cochlear of normal and cisplatin\|treated animals.Results:The expression of Bcl 2 have significant reduction in spiral ganglio and organ of Corti following 6 and 7 days′ CDDP administration respectively,and the expression of Bax have significant increase following 5~7 days′ in the cells spiral ganglio and the organ of Corti of basal turn.Conclusion:The expressions of Bcl 2 and Bax protein have changes under CDDP administration,Bcl 2 family proteins should play an important role in regulation of cochlear cells apoptosis.
