摘要
作者以短期液体培养方法,通过细胞活力测定、形态分类、酵母多糖吞噬试验、NBT还原试验及非特异性酯酶染色等指标,观察了土贝母提取物——结晶E(ST-E)对人早幼粒白血病细胞(HL-60)的诱导分化作用.结果发现ST-E能诱导HL-60细胞向类似成熟中性粒细胞方向分化,细胞形态变化显著,核浆比例变小,核呈肾形或杆状,核染色质浓缩增粗,核仁消失,胞浆嗜天青颗粒消失,还原NBT和吞噬酵母多糖能力显著增强,非特异性酯酶染色阴性,提示ST-E可能是一种新的抗白血病有效药物.
The extract of bolbostemma paniculatum (Maxion) franquet-crystal-E (ST-E) induced differentiation ofhuman promyelocytic leukemia cells (HL-60) into mature-like granulocytes in liquid culture. The cells became closely resembled to normal metamyelocytes or banded and segmented neutrophils, with increased amount cytoplasm and loss of azurophilic granule. ST-E caused significant increase in their ability to phagocytize zymosan particles and to reduce nitroblue tetrazolium (NBT). The cell's staining for a-nephthyl AS-D acetate (nonspecific) esterase was uniformly negative. The above findings suggest that ST-E may be an effective antileukemic drug.
