摘要
作者用乙肝型肝炎病毒表面抗原单克隆抗体(Ab_1)免疫新西兰兔,制备抗独特型抗体(Ab_2).Ab_2经50%饱合硫酸胺沉淀,并通过BALB/c小鼠全血清-sepharose-4B免疫亲和柱,以去除兔抗鼠抗体,经ELISA夹心法及阻断试验,证明Ab_2特异性良好.以Ab_2与正常兔IgG免疫两组BALB/c小鼠,免疫完成后鼠血清以RIA测定两组抗-HBs效价。Ab_2组明显高于正常兔IgG组(t=6.99,P<0.01).提示用Ab_2制备抗独特型疫苗的可能性。
Anti-idiotypic antibody (Ab2) was prepared by immunization of rabbits with monoclonal anti-HBs (Ab1) purified by affinity chromatography. The sandwich ELISA and obstruction ELISA were established for the detection and specific of Ab2. The results show that the Ab2 is specific for monoclonal anti-HBs. Two groups of BALB/c mice were immuned by the Ab, and normal rabbit IgG. The results showed that the Ab2 could induce anti-HBs immune response. So, to develop such a vaccine may be useful against hepatitis B virus.
