摘要
作者采用通用分析型高效弱阴离子交换柱TSK DEAE-5PW,建立了半制备级纯化肾综合征出血热病毒血凝素单克隆抗体的高效液相色谱方法.该法纯化过程仅用3种试剂,采用二元梯度洗脱,一次纯化周期仅为60 min,每次进样处理蛋白质总量可达20 mg,IgG_1的回收率为67%~73%,纯度≥91%,免疫学活性检测获得满意结果,纯化倍数为49.6~51.6。该法对于纯化其它IgG类单克隆抗体具有实际应用价值。
The author describes a high performance liquid chromatography for purification of monoclonal antibodies from mouse ascites liquid. A conventional analytical weak anion exchange column TSK DEAE-5PW can be used with Tris-HAc/NaAc stepwise ionic strength gradient elution. Each time, the total protein sample loading is 20 mg on a semi-preparative scale in an operating cycle of 60 min. The recovery rate of purified IgG1 is 67% -73% with a purity over 91%. The immunological activities of purified IgG, are 49.6 - 51.6 times more than those of crude ascites liquid. The method offers a practical mean for purification of monoclonal antibodies of other IgG types.
