摘要
目的 探讨重庆地区人群肾素基因 (ACAG)n串联重复顺序多态性与原发性高血压的关系。方法 用多聚酶链反应直接扩增肾素基因第 7内含子的四核苷酸 [ACAG]串联重复顺序 ,变性聚丙烯酰胺凝胶电泳结合银染的方法对 80例正常人和 83例原发性高血压患者的基因型进行检测。结果 发现 5种等位基因 ,基因型中以 4 / 4型 (6 2 .6 % ) ,等位基因中以 4型 (77.6 % )最为多见。基因型频率分布符合Hardy Weinberg平衡定律。正常组和高血压组的基因型频率和等位基因频率均无显著性差异 (χ2=3.2 73,P >0 .0 5 ;χ2 =1.2 12 ,P >0 .0 5 )。
Objective To investigate the association of renin gene (ACAG)n short tandem repeat polymorphism (STRP) with essential hypertension in Chongqing. Methods Polymerase chain reaction (PCR) technique combined with silver staining on denaturing DNA sequencing gel was applied to genotype the (ACAG)n STRP located in the 7th intron of renin gene among 80 normotensive and 83 hypertensive subjects. Results Five alleles were discovered in this research. The most frequent genotype and allele of renin gene (ACAG)n STRP are genotype 4/4(62.6%) and allele 4(77.6%).The genotype frequency distribution is in Hardy Weinberg equilibrium. The frequencies of genotype and allele showed no significant difference between two groups(χ 2=3.273, P >0.05;χ 2=1.212, P >0.05). Conclusion Renin gene is not a major contributor to the genetic bases for essential hypertension in Chongqing.
