神经干细胞分化前后基因表达变化的DNA微阵列分析 被引量:2

摘要 目的 了解神经干细胞分化前后的差异表达基因 ,为神经干细胞的分化调控提供基础研究资料。方法 利用细胞培养、DNA微阵列技术等 ,检测了神经干细胞分化前后的差异表达基因。结果 检测到了 10 0 0多个差异表达基因 ,其中明显上调的基因 13 8个 ,明显下调的基因 179个 ,基因种类很多如细胞周期相关基因、发育与分化相关基因、受体与通道相关基因、细胞骨架相关基因、生长因子相关基因、原癌基因和抑癌基因、转录和翻译调节基因、ESTs、代谢调节、蛋白运输相关基因等。结论 大量基因参与了神经干细胞的分化调控 ,其确切调控机制 。 Objective To observe the differential gene expression of neural stem cells(NSCs) in their neurosphere state with differentiation, so as to provide basic research data for differentiation regulation of NSCs. Methods Differential gene expression before and after the differentiation of neural stem cells was investigated by cell culture, DNA microarray, etc. Results More than 1000 genes(138 up regulation genes and 179 down regulation genes) of differential expression were detected. They were related with cell cycle genes, development and differentiation genes, receptor and signal channel genes, cellular skeleton genes, cell signal genes, growth factor genes, oncogenes and anti oncogenes, transcription and translation genes, ESTs genes, metabolic regulation and protein traffic genes, etc. Conclusion A lot of genes are involved in differentiation regulation of neural stem cells, but further studies of the mechanism are needed.
