二次脑损伤后大鼠脑皮层mGluR2 mRNA的变化

摘要 目的 研究弥漫性脑损伤 (diffusebraininjury,DBI)及其合并二次脑损伤 (secondarybraininsults,SBI)后大鼠脑皮层代谢性谷氨酸受体 2亚型 (mGluR2 )的变化及其意义 .方法 SD大鼠 14 0只随机分为正常对照、假手术、DBI与DBI合并SBI4组 .在Marmarou弥漫性脑损伤模型的基础上 ,制成SBI模型 ,于伤后 1,3,6 ,12 ,2 4 ,4 8和 72h进行HE染色和mGluR2mRNA的原位杂交研究 .结果 与正常对照组相比 ,假手术组阳性神经元数无明显改变 (P >0 0 5 ) ;DBI组脑皮层mGluR2mRNA在伤后 12h表达开始减少 ,4 8h降至最低 (P <0 0 5 ) ;DBI合并SBI组mGluR2mRNA表达于伤后 6h即开始减少 ,2 4h降至最低 (P <0 0 5 ) .结论 在DBI及其合并SBI过程中 ,mGluR2mRNA表达降低 .mGluR2参与了DBI和SBI的病理生理过程 ,可能与脑保护有关 . AIM To study the changes and effects of metabotropic glutamate receptor subtype 2 (mGluR2) in diffuse brain injuries (DBI) coupled with secondary brain insults (SBI) in rats. METHODS One hundred and forty SD rats were randomized into four groups: normal control, sham operated, DBI alone and DBI coupled with SBI. The SBI model was made on the basis of Marmarou model. The brain sections were stained with H&E and the mRNA of mGluR2 was detected in 1, 3, 6, 12, 24, 48 and 72 hours after injuries by hybridization in situ. RESULTS Statistical analysis showed no significance between normal control group and sham-operated group ( P >0 05).The number of mGluR2 positive neurons decreased at 12 hours after injury and the biggest drop occurred at 48 hours after injury in the injured cerebral cortex in head injury alone group. However, in head injury with SBI group, there was a significant decrease of the number of mGluR2 positive neurons at 6 hours after injury and the biggest drop occurred at 24 hours after injury. CONCLUSION mGluR2, a factor with the function of protecting brain, may play an important role in the pathophysiology of DBI and SBI.
