基于TNF-α信号通路探究中医药治疗抽动障碍的研究进展

摘要 抽动障碍(TD)是一种常见于儿童和青少年的神经精神性疾病,其发病与神经炎症反应及免疫调节异常密切相关,主要表现为不自主的运动性和发声性抽动。近年来,TNF-α炎症信号通路在TD的病理机制中起重要作用,参与调节神经炎症反应、影响神经元兴奋性及突触传递过程。中医药通过整体调节和多靶点干预,针对TNF-α信号通路中的多个关键环节进行调控,从而减轻神经炎症、抑制过度免疫应答并改善神经功能。本文系统综述中医药通过调控TNF-α炎症信号通路治疗TD的研究进展,重点探讨其在抑制TNF-α介导的神经炎症、调节胶质细胞活化及神经免疫微环境等方面的作用机制,揭示中医药在调节神经递质平衡、保护神经元及改善行为症状等方面的综合调控优势,为阐释中医药治疗TD的现代药理机制提供理论依据,并为靶向TNF-α信号通路的中医药创新研究提供思路与方向。 Tic disorder(TD)is a common neurological and psychiatric disorder in children and adolescents,its onset is closely related to neuroinflammatory response and immune regulation abnormalities,mainly manifested as involuntary motor and vocal tics.In recent years,TNF-αinflammatory signaling pathway has played an important role in pathological mechanism of TD,participating in regulating neuroinflammatory responses,affecting neuronal excitability,and synaptic transmission processes.Traditional Chinese medicine and pharmacy regulates multiple key links in TNF-αsignaling pathway through holistic regulation and multi-target intervention,thereby reducing neuroinflammation,inhibiting excessive immune response,and improving neurological function.This article systematically reviews research progress on traditional Chinese medicine and pharmacy in the treatment of TD by regulating TNF-αinflammatory signaling pathway,focuses on exploring its action mechanism in inhibiting TNF-αmediated neuroinflammation,regulating glial cell activation,and neuroimmune microenvironment,reveals comprehensive regulatory advantages of traditional Chinese medicine and pharmacy in regulating neurotransmitter balance,protecting neurons,and improving behavioral symptoms,in order to provide theoretical basis for explaining modern pharmacological mechanism of traditional Chinese medicine and pharmacy in the treatment of TD,and provide ideas and directions for innovative research in traditional Chinese medicine and pharmacy targeting TNF-αsignaling pathway.
