基于UPLC-MS/MS指纹图谱及化学模式识别对比研究8种紫堇属药材

Comparison of eight Corydalis based on UPLC-MS/MS fingerprint and chemical pattern recognition

摘要 目的揭示8种紫堇属药材中生物碱的差异,为其鉴别提供依据。方法采用超高效液相色谱-三重四极杆质谱联用(UPLC-MS/MS)法测定紫堇属药材中紫堇灵、原阿片碱、延胡索乙素、盐酸小檗碱、比枯枯灵、黄藤素含量。结合聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘法-判别分析(OPLS-DA)比较不同紫堇属药材的差异。结果通过相似度分析发现,不同种紫堇属药材相似度差异较大。HCA、PCA和OPLS-DA将样品分为7类,但无法区分黄堇与珠果黄堇样品。通过OPLS-DA中的变量重要性投影(VIP)筛选出3个差异性标志物:原阿片碱、紫堇灵、盐酸小檗碱。结论本研究建立了一种紫堇属药材UPLC-MS/MS鉴定方法,该方法合理、准确,能有效地鉴别6种紫堇属药材。然而,本研究仍存在一定的局限性,如黄堇与珠果黄堇无法区分。 Objective To compare alkaloids in eight Corydalis and provide a basis for their identification.Methods The contents of corynoline,protopine,tetrahydropalmatine,berberine hydrochloride,bicuculline,and palmatine in Corydalis samples were determined with ultra-performance liquid chromatography coupled with tandem quadrupole mass spectrometry(UPLC-MS/MS).Hierarchical cluster analysis,principal component analysis and orthogonal partial least square discriminant analysis were used to compare differences among the samples.Results Similarity analysis showed distinct differences among the samples.Hierarchical cluster analysis,principal component analysis and orthogonal partial least squares discriminant analysis grouped the samples into seven categories,but failed to distinguish Corydalis pallida from Corydalis speciosa.Three differential markers(protopine,corynoline,and berberine hydrochloride)were identified by orthogonal partial least squares discriminant analysis based on variable importance in projection(VIP)values.Conclusion The study successfully establishes a reliable and accurate UPLC-MS/MS method for the identification of six Corydalis,providing a valuable reference for their differentiation and authentication,but fails to distinguish between Corydalis pallida and Corydalis speciosa.
