摘要
采用基因组挖掘技术,在链霉菌Streptomyces sp.YINM00050的基因组中发现了一个二型PKS基因簇,与苯并异色烷醌类化合物Alnumycin的生物合成基因簇具有较高的相似性.采用多种培养基发酵野生型YINM00050菌株,没有获得该类化合物.通过导入全局调控质粒pSH152-tRNA-Asp-AUC,构建了突变菌株YINM00050-tRNA.对突变株进行发酵,对苯并异色烷醌类化合物进行有针对性的分离,发现了1个新的抗肿瘤活性化合物Alnumycin E.
Through genome mining,a type II polyketide synthase(PKS)gene cluster was identified within the genome of Streptomyces sp.YINM00050.This gene cluster exhibits a high degree of similarity to the biosynthetic gene cluster responsible for the production of alnumycin,a benzoisochromanequinone(BIQ)compound.Nevertheless,no BIQs were isolated from the fermentations of the wild-type strain YINM00050 using various media.Subsequently,the global regulatory plasmid pSH152-tRNA-Asp-AUC was introduced into the wild-type strain,thereby generating the mutant strain YINM00050-tRNA.The subsequent fermentation of this mutant strain enabled the targeted isolation of BIQs.Moreover,a novel anti-tumor compound,alnumycin E,was identified.
