摘要
Two new glutarimide derivatives,2-hydroxy-3-[2-(2-hydroxy-3-methylphenyl-5-hydroxymethyl)-2-oxoethyl]glutarimide(1)and 2-hydroxy-3-[2-[2-hydroxy-3-hydroxymethyl-5-methylphenyl]-2-oxoethyl]glutarimide(2),along with 8 known compounds were isolated from the liquid fermentation broth of endophytic Streptomyces sp.YINM00048 obtained from Agrimonia pilosa.Their structures were elucidated through comprehensive spectroscopic analysis,including nuclear magnetic resonance,high-resolution mass spectrometry,and single-crystal X-ray diffraction,supported by time-dependent density functional theory(TDDFT)-electronic circular dichroism(ECD)calculations.Furthermore,compound 2 displayed weak cytotoxic activity against A549 and SMMC-7721 cell lines with IC50 values of 62.69 and 55.07μmol/L,respectively.
通过对药用植物仙鹤草(Agrimonia pilosa)内生链霉菌YINM00048的液体发酵产物的系统探究,共分离获得10个化合物,包括2个结构新颖的戊二酰亚胺类衍生物2-羟基-3-[2-(2-羟基-3-甲基-5-羟甲基苯基)-2-氧代乙基]戊二酰亚胺(1)和2-羟基-3-[2-(2-羟基-3-羟甲基-5-甲基苯基)-2-氧代乙基]戊二酰亚胺(2).通过核磁共振(1D/2D NMR)、高分辨质谱(HR-ESI-MS)、单晶X射线衍射等技术的综合应用,结合含时密度泛函理论-电子圆二色谱(TDDFT-ECD)计算法,确证了化合物1和2的平面结构和绝对构型.体外活性评价显示,化合物2对人肺癌细胞A549和人肝癌细胞SMMC-7721具有较弱的抑制活性,IC50值分别为62.69和55.07μmol/L.
