摘要
为建立黑米稻YH205的高效离体再生体系,以黑米稻YH205成熟胚为外植体,系统优化了其组织培养的各个关键环节。结果表明,最佳消毒方案为75%酒精90 s+0.1%HgCl_(2) 8 min,污染率17.50%,诱导率68.33%。在N6培养基中添加5.5 mg/L 2,4-D+1.0 mg/L 6-BA时诱导率达89.17%;1.2 mg/L TDZ+3.0 mg/L 6-BA培养21 d增殖率达439.97%。在NB培养基添加0.3 mg/L NAA+1.5 mg/L KT时分化率为73.33%。在1/2 MS培养基中添加0.5 mg/L NAA生根效果最佳。
In order to establish an efficient in vitro regeneration system for black rice(Oryza sativa L.var.nigra)variety YH205,the mature embryos of black rice YH205 were used as explants.The key steps of tissue culture were systematically optimized.The results showed that the optimal disinfection scheme was 90 seconds of 75%alcohol+8 minutes of 0.1%HgCl_(2) ,with a contamination rate of 17.50%and an induction rate of 68.33%.When 5.5 mg/L 2,4-D+1.0 mg/L 6-BA was added to the N6 medium,the induction rate reached 89.17%.After 21 days of cultivation with 1.2 mg/L TDZ+3.0 mg/L 6-BA,the proliferation rate reached 439.97%.When 0.3 mg/L NAA+1.5 mg/L KT was added to the NB medium,the differentiation rate was 73.33%.The rooting effect was the best when 0.5 mg/L NAA was added to the 1/2 MS medium.
