摘要
目的:探讨Sirt5通过调控Ucp 1表达对白色脂肪组织(WAT)褐变改善肥胖的影响。方法:将30只C57BL/6J雄性小鼠随机分为对照组、模型组和Sirt 5过表达组,每组10只。对照组进行正常饮食,模型组和Sirt 5过表达组进行高脂饮食构建肥胖模型。进行糖耐量和胰岛素耐量检测,采用RT-PCR检测Sirt 5 mRNA相对表达水平,通过HE染色检测小鼠肝组织和WAT中的脂质堆积情况,进行组织称重和TG、TC水平检测,采用免疫组化染色检测Ucp1蛋白。结果:与对照组相比,模型组小鼠WAT中Sirt 5 mRNA相对表达及Ucp1蛋白阳性面积百分比均明显降低(P<0.05);与模型组相比,Sirt 5过表达组小鼠WAT中Sirt 5 mRNA相对表达及Ucp1蛋白阳性面积百分比显著升高(P<0.05)。与对照组相比,模型组小鼠的肝组织和WAT的TG、TC水平及肝组织和WAT的重量明显升高(P<0.05);与模型组相比,Sirt 5过表达组小鼠的肝组织和WAT的TG、TC水平及肝组织和WAT的重量显著降低(P<0.05),小鼠肝组织和WAT中的脂质堆积明显得到改善(P<0.05)。结论:Sirt 5过表达可以调节脂质水平,减轻肥胖相关症状,促进了WAT的褐变,从而为肥胖的干预提供了新的靶点。
Objective:To explore the effect of Sirt5 on the browning of white adipose tissue(WAT)and its impact on obesity by regulating Ucp 1 expression.Methods:Thirty male C57BL/6J mice were randomly divided into a control group,a model group,and a Sirt 5 overexpression group,with 10 mice in each group.The control group was given a normal diet,while the model group and Sirt 5 overexpression group were fed a high-fat diet to establish an obesity model.Glucose tolerance and insulin tolerance tests were conducted,RT-PCR was used to detect the relative expression level of Sirt 5 mRNA,HE staining was performed to examine lipid accumulation in the liver(WAT)and adipose tissue(WAT)of the mice,tissue weighing and TG,TC level detection were carried out,and immunohistochemistry staining was used to detect Ucp1 protein.Results:Compared with the control group,the expression of Sirt 5 mRNA and the percentage of positive area of Ucp1 protein in the WAT of the model group mice were significantly reduced(P<0.05);compared with the model group,the Sirt 5 overexpression group mice showed a significant increase in the percentage of positive area of Sirt 5 mRNA and Ucp1 protein in the WAT(P<0.05).Compared with the control group,the levels of TG and TC in the liver tissue and WAT of the model group mice,as well as the weight of liver tissue and WAT,were significantly increased(P<0.05);compared with the model group,the Sirt 5 overexpression group mice showed a significant decrease in the levels of TG and TC in the liver tissue and WAT,as well as the weight of liver tissue and WAT(P<0.05),the lipid accumulation in the liver and adipose tissue of the mice was significantly improved(P<0.05).Conclusions:Overexpression of Sirt 5 can regulate lipid levels,alleviate obesity-related symptoms,and promote the browning of WAT,thus providing a new target for the intervention of obesity.
