摘要
目的研究卵巢癌组织中F-box蛋白31(FBXO31)、Prospero相关同源盒蛋白1(PROX1)表达与患者病理特征、预后的相关性,为患者的病理风险评估、预后预测及潜在治疗靶点挖掘提供理论依据。方法选取2019年1月至2021年6月本院收治的卵巢癌患者138例,根据是否死亡分为死亡组(n=59)、存活组(n=79)。利用免疫组化法检测FBXO31、PROX1表达;采用Cox回归筛选患者死亡的影响因素;利用Kaplan-Meier分析FBXO31、PROX1表达对卵巢癌患者生存率的影响。结果卵巢癌组织中FBXO31阴性表达、PROX1阳性表达的比例较高(χ^(2)=50.274、83.781,P<0.05)。在低分化、有淋巴结转移以及TNM分期为Ⅲ~Ⅳ期的患者中,FBXO31的阴性表达比例更高(χ^(2):4.524~12.644,P<0.05),而PROX1的阳性表达比例更高(χ^(2):6.072~10.107,P<0.05)。卵巢癌患者的预后与年龄、肿瘤大小、病理分型无关(P>0.05),而低分化(HR=1.756,95%CI:1.212~2.543)、淋巴结转移(HR=2.122,95%CI:1.398~3.221)、TNMⅢ~Ⅳ期(HR=2.472,95%CI:1.590~3.842)、FBXO31阴性(HR=2.583,95%CI:1.735~3.845)以及PROX1阳性(HR=2.044,95%CI:1.274~3.278)均与死亡风险升高相关(P<0.05)。FBXO31阴性表达生存率49.09%低于阳性89.29%(P<0.05),PROX1阳性表达生存率48.57%低于阴性84.85%(P<0.05)。结论卵巢癌组织中FBXO31、PROX1表达与卵巢癌病理特征有关,FBXO31、PROX1是卵巢癌患者预后的影响因素。
Objective To investigate the expression of F-box protein 31(FBXO31)and prospero-related homeobox 1(PROX1)in ovarian cancer tissues and to analyze their correlations with pathological characteristics and prognosis,so as to provide a theoretical basis for pathological risk assessment,prognostic prediction,and the identification of potential therapeutic targets in ovarian cancer patients.Methods A total of 138 patients with ovarian cancer treated in our hospital from January 2019 to June 2021 were enrolled.According to survival status,patients were divided into a deceased group(n=59)and a survival group(n=79).The expression of FBXO31 and PROX1 was detected by immunohistochemistry.Cox proportional hazards regression analysis was used to identify factors influencing mortality.Kaplan-Meier survival analysis was performed to evaluate the effects of FBXO31 and PROX1 expression on overall survival in ovarian cancer patients.Results The proportions of negative FBXO31 expression and positive PROX1 expression were relatively high in ovarian cancer tissues(χ^(2)=50.274 and 83.781,respectively,P<0.05).Among patients with poorly differentiated tumors,lymph node metastasis,or TNM stage Ⅲ-Ⅳ,the proportion of negative FBXO31 expression was higher(χ^(2):4.524-12.644,P<0.05),while the proportion of positive PROX1 expression was higher(χ^(2):6.072-10.107,P<0.05).The prognosis of ovarian cancer patients was not associated with age,tumor size,or pathological type(P>0.05).Poor differentiation(HR=1.756,95%CI:1.212-2.543),lymph node metastasis(HR=2.122,95%CI:1.398-3.221),TNM stage Ⅲ-Ⅳ(HR=2.472,95%CI:1.590-3.842),negative FBXO31 expression(HR=2.583,95%CI:1.735-3.845),and positive PROX1 expression(HR=2.044,95%CI:1.274-3.278)were all associated with an increased risk of mortality(P<0.05).The survival rate for negative FBXO31 expression was 49.09%,lower than the 89.29% for positive expression(P<0.05).The survival rate for positive PROX1 expression was 48.57%,lower than the 84.85% for negative expression(P<0.05).Conclusion The expressions of FBXO31 and PROX1 in ovarian cancer tissues are associated with the pathological features of ovarian cancer.FBXO31 and PROX1 are influential factors in the prognosis of ovarian cancer patients.
