摘要
目的探讨ADP核糖基化因子的GTP酶激活蛋白1(ASAP1)、肌动蛋白束蛋白1(FSCN1)在胃癌中的表达、预后相关性。方法运用UALCAN、Kaplan-Meier数据库分析ASAP1、FSCN1在胃癌组织与正常胃黏膜组织中的表达与患者预后的关系,并采用qRT-PCR、免疫组化验证ASAP1、FSCN1在胃癌与癌旁的表达差异。结果与正常胃黏膜组织相比,ASAP1、FSCN1在胃癌组织中表达升高(P<0.01),且ASAP1、FSCN1高表达组患者的总生存率下降(P<0.05)。qRT-PCR显示胃癌中ASAP1、FSCN1 mRNA的表达水平高于癌旁正常黏膜组织(P<0.05)。免疫组化结果显示,胃癌ASAP1、FSCN1阳性表达率高于癌旁正常胃黏膜组织,差异均有统计学意义(P<0.05)。ASAP1 mRNA和FSCN1 mRNA的表达成正相关(r=0.558,P=0.000)。ASAP1、FSCN1的表达与胃癌浸润深度、淋巴结转移及远处转移相关(P<0.05)。Kaplan-Meier生存分析发现,ASAP1、FSCN1阴性表达总体生存率高于阳性表达(P<0.05)。结论ASAP1、FSCN1在胃癌组织中均高表达,2者可能协同促进胃癌的发生和发展,且在胃癌的浸润、转移中发挥了重要作用。
Objectivee This paper aims to investigate the expression of ADP ribosylation factor GTP-activating protein 1(ASAP1)and actin bundle protein 1(FSCN1)in gastric cancer and its correlation with prognosis.Methods UALCAN and Kaplan-Meier databases were used to analyze the expression differences of ASAPI and FSCN1 in gastric cancer and normal gastric mucosa and their relationship with prognosis.Furthermore,the expression dfferences of ASAPl and FSCN1 in gastric cancer and adjacent tissues were verified by qRT-PCR and immunohistochemistry.Results Compared with normal gastric mucosa,the levels of ASAP1 and FSCN1 were increased in gastric cancer(P<0.05),and the overall survival rate of patients with high expression of ASAP1 and FSCN1 decreased significantly(P<0.05).qRT-PCR showed that mRNA levels of ASAP1 and FSCN1 in gastric cancer were higher than those in normal mucosa adjacent to cancer,with the differences statistically significant(P<0.05).Immunohistochemical results showed that the positive rates of ASAPl and FSCN1 were higher than in normal gastric mucosa adjacent to cancer(P<0.05).Expression of ASAPI mRNA and FSCN1 mRNA was positively correlated(r=0.558,P=0.000).In addition,ASAP1 and FSCN1 was correlated with the depth of invasion,lymph node metastasis and distant metastasis of gastric cancer(P<0.05).Kaplan-Meier analysis showed that the overall survival rate of negative ASAP1 and FSCN1 expression was higher than that of positive expression(P<0.05).Conclusion ASAPI and FSCNI are highly expressed in gastric cancer tissues,they may synergistically promote the occurrence and development of gastric cancer,and may play an important role in the invasion and metastasis of gastric cancer.
