摘要
目的:通过建立过氧化氢(H_(2)O_(2))对骨骼肌肌肉母细胞(SOL8)的氧化损伤模型,从细胞水平探讨苦荞低聚肽对SOL8增殖细胞氧化损伤的预保护作用,并探讨其可能机制。方法:(1)CCK-8法测定72 h内不同浓度苦荞低聚肽对SOL8细胞活力的影响,筛选苦荞低聚肽的安全及最佳剂量。(2)测定不同浓度H_(2)O_(2)对SOL8增殖细胞活力的影响,确定H_(2)O_(2)氧化应激模型建立的最佳剂量和作用时间。(3)探讨苦荞低聚肽对SOL8增殖细胞氧化应激损伤的预保护作用;(4)检测干预后细胞中总抗氧化能力(T-AOC)、乳酸脱氢酶(LDH)活力、过氧化氢酶(CAT)活力及丙二醛(MDA)含量,并使用DHE荧光探针和DCFH-DA探针检测SOL8细胞中活性氧产生情况。结果:(1)苦荞低聚肽浓度在1~20 mg/mL时,细胞活力随干预时间逐步升高,综合考虑干预时间和浓度,选取10 mg/mL浓度进行后续实验,作用时间48 h。(2)0.05 mmol/L的H_(2)O_(2)作用于SOL8细胞1 h,CCK-8值极显著低于对照组(P<0.001)。(3)干预组的ROS产生量显著降低(P<0.001)。(4)干预组与过氧化氢组相比:总抗氧化能力(T-AOC)提高9.4%、乳酸脱氢酶(LDH)活力极显著上升(P<0.001)、过氧化氢酶(CAT)活力显著升高(P<0.001),脂质过氧化产物(MDA)含量降低3%。结论:苦荞低聚肽对H_(2)O_(2)损伤的SOL8细胞具有预保护作用,可能与降低活性氧攻击及增强胞内酶活力维持了细胞完整性有关。
【Objective】By establishing a model of oxidative damage of hydrogen peroxide(H_(2)O_(2))on skeletal muscle muscle blasts(SOL8),the preprotective effect and the potential mechanism of Tartary buckwheat oligopeptide on the proliferation of SOL8 cells damaged by H_(2)O_(2)-induced oxidative stress was investigated at the cellular level.【Method】CCK-8 method was used to determine the effect of tartary buckwheat oligopeptide on the viability of SOL8 cells within 72 h,and to screen the safety and optimal dose of tartary buckwheat oligopeptide.The effect of different concentrations of H_(2)O_(2) on the viability of SOL8 proliferating cells was evaluated.The preprotective effect of tartary buckwheat oligopeptide on oxidative stress damage in SOL8 proliferating cells was investigated.The total antioxidant capacity(T-AOC)of cells was detected after intervention.Lactate dehydrogenase(LDH)activity,catalase(CAT)activity and malondialdehyde(MDA)content were determined,and DHE fluorescent probe and DCFH-DA probe were used to determine the production of reactive oxygen species(ROS)in SOL8 cells【.Result】When the concentration of tartary buckwheat oligopeptide was 1—20 mg/mL,the cell viability gradually increased,when the concentration of tartary buckwheat oligopeptide was 10 mg/mL,it could promote cell proliferation.The concentration of 10 mg/mL of tartary buckwheat oligopeptide was selected for follow-up experiments,and the effect time was 48 h.0.05 mmol/L H_(2)O_(2) acted on SOL8 cells for 1 h,and the CCK-8 value was significantly lower than that of the control group(P<0.001).The ROS in the intervention group was significantly reduced(P<0.001).Compared with hydrogen peroxide,the total antioxidant capacity(T-AOC)increased by 9.4%,the activity of lactate dehydrogenase(LDH)was significantly increased(P<0.001),the activity of catalase(CAT)was significantly increased(P<0.001),while the content of lipid oxidation(MDA)decreased by 3%.【Conclusion】Tartary buckwheat oligopeptide has significant preprotective effect on H_(2)O_(2)-damaged SOL8 cells,and the mechanism may be related to the simultaneous enhancement of antioxidant capacity,intracellular enzyme activity,reduction of reactive oxygen species attack and maintenance of cell integrity.
