锁阳鞣质对D-半乳糖致衰老模型小鼠抗氧化与抗炎作用的研究

Antioxidant and Anti-Inflammatory Effects of tannins from Suoyang(Cynomorium songaricum Rupr)on the Aging Model Mice Induced by D-galactose

摘要 目的研究锁阳鞣质对D-半乳糖致衰老小鼠体内抗氧化、抗炎作用的影响。方法60只C57BL/6J小鼠随机分为空白对照组、模型组、阳性对照组,锁阳鞣质高剂量组[200 mg/(kg·d)]、中剂量组[100 mg/(kg·d)]、低剂量组[50 mg/(kg·d)]。对照组以外的小鼠均通过颈背部皮下注射D-半乳糖[150 mg/(kg·d)],连续8周建立衰老模型,阳性对照组与锁阳鞣质给药组灌胃干预8周,对照组与模型组以同等方法给予等体积生理盐水。实验8周后,检测各组小鼠血清及肝脏、肾脏及脾脏中丙二醛(malondialdehyde,MDA)含量和超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)等酶活力,并测定小鼠血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)及白细胞介素-18(interleukin-18,IL-18)的含量。结果锁阳鞣质可显著降低小鼠血清和肝脏、肾脏和脾脏中MDA含量,提高SOD、GSH-Px及CAT活力,同时还可降低小鼠血清中TNF-α、IL-6、IL-1β及IL-18等促炎因子水平。结论锁阳鞣质可以提高D-半乳糖衰老模型小鼠的抗氧化和抗炎能力,可能通过抑制体内氧化应激及提高机体抗炎因子水平发挥延缓衰老的作用。 Objective To study the antioxidant and anti-inflammatory effects of Suoyang(Cynomorium songaricum Rupr)tannin on D-galactose-induced aging mice.Methods Sixty of C57BL/6J mice were randomly divided into blank control group,model group,positive control group,high[200 mg/(kg·d)],medium[100 mg/(kg·d)]and low[50 mg/(kg·d)]dose groups.The mice except the control group were injected subcutaneously with D-galactose[150 mg/(kg·d)]in the neck and back,and the aging model was established for 8 consecutive weeks.The positive control group and the cynotannin administration group were administrated by gavage for 8 weeks,and the control group and the model group were administrated with the equal volume of normal saline in the same way.After 8 weeks of the experiment,the content of malondialdehyde(MDA)and the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and catalase(CAT)in serum,liver,kidney and spleen of mice in each group were detected.The serum levels of tumor necrosis factor(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β)and interleukin-18(IL-18)were measured.Results The contents of MDA in serum,liver,kidney and spleen of mice were significantly decreased,the activities of SOD,GSH-Px and CAT were increased,and the levels of TNF-α,IL-6,IL-1βand IL-18 in serum of mice were also decreased.Conclusion Suoyang(Cynomorium songaricum Rupr)tannin can improve the antioxidant and anti-inflammatory ability of D-galactose aging model mice,and may play a role in delaying aging by inhibiting oxidative stress and improving the levels of anti-inflammatory factors in the body.
