犬尿喹啉酸调节血管平滑肌细胞的表型转换和凋亡治疗腹主动脉瘤相关的血管损伤

Kynurenic acid treats vascular injury associated with abdominal aortic aneurysms by regulating phenotypic transform and apoptosis of vascular smooth muscle cells

摘要 目的 观察犬尿喹啉酸(KynA)对腹主动脉瘤(AAA)小鼠模型的治疗效果和作用机制。方法 血管紧张素II(Ang II)诱导小鼠发生AAA,将健康雄性ApoE^(–/–)小鼠随机分为假手术组(Sham)、模型组(AAA)和治疗组(AAA+KynA),采用HE染色观察小鼠血管平滑肌细胞(VSMC)的形态变化,TUNEL染色检测VSMC的凋亡情况,免疫荧光检测α-SMA蛋白的表达,Western blot方法检测VSMC中与表型转换和凋亡相关的蛋白表达水平。结果 KynA能降低小鼠的AAA发生率,上调收缩型VSMC标志物α-SMA、SM22α的表达,抑制合成型VSMC标志物骨桥蛋白(OPN)的表达,上调Bcl-2的表达,抑制Bax的表达,抑制VSMC的凋亡。结论 KynA能调节VSMC由合成型到收缩型的转换,并抑制VSMC凋亡。 Objective To observe the therapeutic effect and molecular mechanism of kynurenic acid(KynA)on mice model of abdominal aortic aneurysm(AAA).Methods AngiotensinⅡ(AngⅡ)induces abdominal aortic aneurysm(AAA)in mice.Healthy male ApoE-/-mice were randomly divided into sham group(Sham),model group(AAA),and treatment group(AAA+KynA).HE staining was used to observe the morphological changes of vascular smooth muscle cells(VSMC).TUNEL staining was used to detect the apoptosis of VSMC.The immunofluorescence was used to detect the expression ofα-SMA.Western blot was used to detect the expressions of proteins related to phenotype transition and ap-optosis in VSMC.Results KynA could reduce the incidence of AAA in mice,up-regulate the expressions of contractile VSMC markersα-SMA and SM22α,inhibit the expression of synthetic VSMC marker osteopontin(OPN),up-regulate the expression of Bcl-2,inhibit the expression of Bax,and inhibit the apoptosis of VSMC.Conclusion KynA can regu-late the transformation of VSMC from synthetic to contractile and inhibit VSMC apoptosis.
