虫草素介导NRP1调节AFF3增加乳腺癌细胞对他莫昔芬的敏感性

摘要 目的 探究虫草素对乳腺癌细胞他莫昔芬敏感性的影响及机制。方法 MCF-7细胞用不同浓度虫草素处理后,采用平板克隆形成实验和流式细胞术检测细胞对他莫昔芬的敏感性;RT-qPCR和Western blot检测细胞中NRP1 mRNA和蛋白表达;Western blot检测细胞中AFF3蛋白表达。将MCF-7细胞分为Control组、CD组、CD+ov-NC组和CD+ov-NRP1组,并按上述方法进行检测。结果 虫草素剂量依赖降低他莫昔芬处理的乳腺癌细胞的克隆形成数并增加细胞凋亡率,降低乳腺癌细胞中NRP1和AFF3表达;过表达NRP1部分逆转虫草素对他莫昔芬处理的乳腺癌细胞克隆形成数的抑制作用及细胞凋亡的促进作用,过表达NRP1还逆转虫草素对乳腺癌细胞中AFF3表达的下调作用。结论 虫草素通过下调NRP1表达增加乳腺癌细胞对他莫昔芬的敏感性,其机制与降低AFF3表达有关。 Objective To explore the effect and mechanism of cordycepin on the sensitivity of breast cancer cells to tamoxifen.Methods After MCF-7 cells were treated with different concentrations of cordycepin,the sensitivity of MCF-7 cells to tamoxifen was detected by plate clone formation test and flow cytometry.RT-qPCR and Western blot were used to detect the expressions of NRP1 mRNA and protein in cells.The expression of AFF3 in cells was detected by Western blot.MCF-7 cells were divided into control group,CD group,CD+ov-NC group and CD+ov-NRP1 group,and were detected according to the above methods.Results Cordycepin dose-dependently decreased the number of clones and increased the apoptosis rate of breast cancer cells treated with tamoxifen,and decreased the expressions of NRP1 and AFF3 in breast cancer cells.Overexpression of NRP1 partially reversed the inhibition of cordycepin on the number of clones and the promotion of apoptosis rate in tamoxifen-treated breast cancer cells,and also reversed the down-regulation of cordycepin on the expression of AFF3 in breast cancer cells.Conclusion Cordycepin increases the sensitivity of breast cancer cells to tamoxifen by down-regulating the expression of NRP1,and its mechanism is related to the decrease of AFF3 expression.
