摘要
食二酸迟缓乳杆菌(Lentilactobacillus diolivorans,Lent.diolivorans)的模式株分离自青贮饲料,在饲料发酵中具有广泛应用。然而,目前该菌种优良种质资源少,涉及耐酸、抑制霉菌等与应用相关的性状研究较少。文章旨在获得具有优良耐酸特性和抑制霉菌活性的Lent.diolivorans,为青贮饲料提供优良乳酸菌补充剂。文章对分离自乳制品中的254株Lent.diolivorans进行耐酸(pH3.0)筛选,筛选出1株耐受性最好的菌株Lent.diolivorans IMAU13829。通过96孔板法和双层平板法测定其抑制霉菌活性,采用蛋白酶处理和酸处理等方法分析乳酸菌抑制霉菌的有效成分,并进行了形态学特征描述和安全性试验。结果表明,菌株Lent.diolivorans IMAU13829对黑曲霉、娄地青霉、串珠镰刀菌具有显著抑菌活性,推测其抑制霉菌的物质为有机酸类。菌株Lent.diolivorans IMAU13829对卡那霉素具有耐药性,不产生溶血反应。综上所述,文章筛选出1株Lent.diolivorans IMAU13829可用作青贮饲料补充剂,抑制霉菌繁殖,改善青贮品质。
The type strain of Lentilactobacillus diolivorans(Lent.diolivorans)was initially isolated from silage and had been widely used in feed fermentation.However,there are few excellent germplasm resources of this strain,and there are few studies on its application-related traits such as acid-resistance and inhibition of mold.The purpose of this article was to obtain Lent.diolivorans with excellent acid-resistance and antifungal activity,providing excellent lactic acid bacteria supplements for silage.In this article,254 strains of Lent.diolivorans isolated from dairy products were screened for acid-resistance at pH3.0 and selected one strain Lent.diolivorans IMAU13829 with the best tolerance.The antifungal activity was determined by 96-well plate method and double-agar plate method.The effective components of lactic acid bacteria inhibiting molds were analyzed by protease treatment and acid treatment.The morphological characteristics description and safety test were carried out.The results showed that the strain Lent.diolivorans IMAU13829 had significant antifungal activitives against Aspergillus niger,Penicillium roqueforti and Fusarium moniliforme.It was speculated that its antifungal substances were organic acids.IMAU13829 showed resistance to kanamycin.It did not produce hemolytic reactions.In conclusion,this article screened a strain of Lent.diolivorans IMAU13829 that can be used as silage supplements to inhibit mold reproduction and improve silage quality.
