摘要
目的:探讨添加牙骨质蛋白1衍生肽1(CEMP1-p1)多肽对Clinpro^(TM)窝沟封闭剂促进釉质再矿化能力的影响。方法:配制CEMP1-p1多肽浓度分别为1%、4%、8%加入Clinpro^(TM)窝沟封闭剂(依次命名为C1、C4和C8组),比较不同窝沟封闭剂固化后的表面硬度、流动性、界面微渗漏,高效液相色谱定量测定CEMP1-p1多肽的释放,能谱仪分析界面釉质层的元素分布变化。结果:各组窝沟封闭剂固化后表面硬度无明显差异(P>0.05),C8组窝沟封闭剂的流动性低于其他组(P<0.05),且界面微渗漏发生率明显高于其他组(P<0.05)。C4组窝沟封闭剂CEMP1-p1的释放量在72 h内的各个时间段高于C1组(P<0.05),C4组离体牙在人工唾液中孵育24 h后釉质的钙/磷的比值为(1.67±0.09)(P<0.05),与正常釉质的钙/磷的比例接近。结论:添加4%CEMP1-p1不会影响Clinpro^(TM)窝沟封闭剂的机械性能,并且会促进釉质的再矿化。
Objective:Cementum protein 1-derived peptide 1(CEMP1-p1)was added to commercial Clinpro^(TM) pit fissure sealant and the effect of modification on enamel remineralization was compared.Methods:Experimental pit and fissure sealants were formulated with CEMP1-p1 at 1%,4%,and 8%(C1,C4 and C8 groups),and the surface hardness,flow properties and interface micro-leakage of different pit and fissure sealants were measured.High performance liquid chromatography was used to compare the release of peptides;energy dispersive spectrometer was conducted to analyze the elemental distribution changes of the enamel layer at the interface.Results:The surface hardness of pit and fissure sealants in different groups after curing showed no significant difference(P>0.05),the fluidity of the pit and fissure sealant group with 8%CEMP1-p1 was lower than that of the other groups(P<0.05),and the incidence of interfacial leakage rate was significantly higher than that of other groups(P<0.05).The release of CEMP1-p1 in the C4 group was higher than that in the C1 group with in 72 hours at all time points(P<0.05).After incubating in artificial saliva for 24 hours,the Ca/P ratio of enamel in C4 group increased to(1.67±0.09)(P<0.05),approaching the Ca/P ratio of normal enamel.Conclusions:The addition of 4% CEMP1-p1 can promote enamel remineralization without affecting the mechanical properties of Clinpro^(TM) pit and fissure sealant.
