摘要
[目的]探究考布他汀A4(Combretastatin A4,CA4)类似物调控PI3K/AKT通路对胃癌SGC7901细胞增殖、侵袭的影响。[方法]将胃癌细胞系SGC7901细胞随机分为3组:对照组、CA4类似物组和LY294002组。CA4类似物组的SGC7901细胞使用24.18 ng/mL CA4类似物处理24 h,LY294002组的SGC7901细胞使用10μmol/L的LY294002处理24 h。以CCK-8实验分析SGC7901细胞的增殖能力,以Transwell实验分析SGC7901细胞的侵袭能力,以流式细胞术分析SGC7901细胞的细胞周期,以蛋白免疫印迹实验分析PI3K/AKT信号通路蛋白表达。[结果]与对照组相比,CA4类似物组和LY294002组的SGC7901细胞增殖活性下降(P<0.05),侵袭数量显著降低(P<0.05),细胞周期滞于G2/M期的比例增加(P<0.05),PI3K、AKT蛋白表达水平降低(P<0.05)。[结论]CA4类似物能够通过抑制PI3K/AKT信号通路进而减弱胃癌SGC7901细胞的增殖、侵袭活性,并诱导SGC7901细胞的细胞周期滞于G2/M期。
[Objective]To investigate the effect of combretastatin A4(Combretastatin A4,CA4)analogue on the proliferation and invasion of gastric cancer SGC7901 cells by regulating PI3K/AKT pathway.[Method]Gastric cancer cell line SGC7901 cells were randomly divided into 3 groups:control group,CA4 analogue group and LY294002 group.SGC7901 cells in CA4 an-alogue group were treated with 24.18 ng/mL CA4 analogue for 24 h,and SGC7901 cells in LY294002 group were treated with 10μmol/L LY294002 for 24 h.The proliferation ability of SGC7901 cells was analyzed by CCK-8 assay.The invasion ability of SGC7901 cells was analyzed by Transwell assay.The cell cycle of SGC7901 cells was analyzed by flow cytometry.The protein expression of PI3K/AKT signaling pathway was analyzed by Western blot.[Result]Compared with the control group,the growth rate of SGC7901 cells in the CA4 analogue group and LY294002 group decreased(P<0.05).The number of invasive cells was significantly decreased(P<0.05).The proportion of cell cycle arrest in G2/M phase increased(P<0.05).The protein expression of PI3K/AKT was significantly decreased(P<0.05).[Conclusion]The CA4 analogues can inhibit the PI3K/AKT signaling pathway,thereby reducing the proliferation of gastric cancer SGC7901 cells,invasion activity,and induce the cell cycle of SGC7901 cells to be arrested at the G2/M phase.
