摘要
[目的]研究CBX6在鼻咽癌中的表达及其对6-10B细胞上皮间质转化的影响。[方法]随机选取2022年1月-2024年1月保藏的鼻咽癌组织及癌旁组织各18例,将鼻咽癌6-10B细胞随机分3组(3×10^(5)个/组):空白对照组(仅加培养基)、si NC组(转染20μmol/L阴性对照siRNA)、si CBX6组(转染20μmol/L CBX6敲低siRNA)。采用免疫组化、蛋白免疫印迹实验检测鼻咽癌组织和癌旁组织CBX6表达水平,MTT实验检测各组6-10B细胞增殖能力,Transwell实验检测各组6-10B细胞侵袭能力,流式细胞术检测各组6-10B细胞凋亡率,蛋白免疫印迹分析6-10B细胞中Snail/Zeb1通路及上皮间质转化相关蛋白的表达。[结果]鼻咽癌组织中CBX6表达水平高于癌旁组织(P<0.05)。与空白对照组、si NC组的6-10B细胞比较,si CBX6组的6-10B细胞增殖、侵袭能力降低、凋亡率增加、Ncadherin、Vimentin表达下调,E-cadherin表达上调,Snail、Zeb1表达下调(P<0.05)。[结论]下调CBX6能够通过抑制Snail/Zeb1信号通路进而减弱鼻咽癌6-10B细胞的增殖、侵袭活性,促进6-10B细胞凋亡。
[Objective]To investigate the expression of CBX6 in nasopharyngeal carcinoma and its effect on epithelial-mesen-chymal transition of 6-10B cells.[Method]Eighteen cases of nasopharyngeal carcinoma tissues and adjacent tissues each were randomly selected from the biobank of our hospital during the period from January 2022 to January 2024.Nasopharyngeal carcinoma 6-10B cells were randomly divided into three groups(3×10^(5) cells per group):blank control group(only medium was added),si NC group(transfected with 20μmol/L negative control siRNA),and si CBX6 group(transfected with 20μmol/L CBX6 knockdown siRNA).The expression level of CBX6 in nasopharyngeal carcinoma tissues and adjacent tissues was ana-lyzed by immunohistochemistry and Western blot.The proliferation ability of in each group of 6-10B cell was analyzed by MTT experiment.The invasion ability of 6-10B cells in each group was analyzed by Transwell assay.The apoptosis rate of 6-10B cells in each group was analyzed by flow cytometry.Snail/Zeb1 pathways and the expression level of a protein involved in trans-formation of epithelial mesenchymal in 6-10B cells were analyzed by protein immunoblot.[Result]In nasopharyngeal carcino-ma tissue CBX6 expression level higher than the tissue adjacent to carcinoma(P<0.05).Compared with 6-10B cells in blank control group and si NC group,the proliferation and invasion ability of 6-10B cells in si CBX6 group were decreased,the apop-tosis rate was increased,the expression of N-cadherin and Vimentin was decreased,the expression of E-cadherin was in-creased,and the expression of Snail and Zeb1 was decreased.Cut CBX6(P<0.05).[Conclusion]Down-regulation of CBX6 can inhibit the Snail/Zeb1 signaling pathway,thereby weakening the proliferation and invasion activities of nasopharyngeal car-cinoma 6-10B cells and promoting apoptosis of 6-10B cells.
