植物乳杆菌对蛋鸡产气荚膜梭菌体外抑制作用的研究

摘要 本研究从某蛋鸡场患鸡肠道中分离到产气荚膜梭菌SW株,体外评估植物乳杆菌(Lactobacillus plantarum)ZG-7株对蛋鸡源产气荚膜梭菌的抑制作用。通过共培养试验、琼脂扩散法、产酸能力测定以及对其无细胞上清液(CFS)的pH稳定性、酶处理和有机酸成分分析,探究其抑菌活性和作用物质基础。结果显示:共培养体系中SW株在生长4 h后即受到显著抑制,至6 h细菌生长已极为缓慢,LP ZG-7株CFS对SW产生了清晰的抑菌圈(16.4 mm);CFS调整到pH3后的抑菌圈变化不大(16.5 mm),调整到pH7后的抑菌圈直径减少到5.5 mm,表明抑菌活性来源于酸性物质。蛋白酶K处理几乎完全消除了CFS的抑菌活性,胰蛋白酶和胃蛋白酶处理的抑菌圈大小约10 mm,无酶处理的上清液抑菌圈直径约为16 mm,提示植物乳杆菌CFS中存在对蛋白酶敏感的蛋白类抑菌物质。LP ZG-7株在培养6 h内pH值从6.3迅速降至4.0以下,24 h终末pH值约为3.7,表现出强大的产酸能力。综上所述,植物乳杆菌ZG-7通过产酸和分泌细菌素类物质,在体外对蛋鸡源产气荚膜梭菌表现出高效的抑制作用,显示出作为预防坏死性肠炎的潜在益生菌候选株的应用前景。 In this study,Clostridium perfringens strain SW was isolated from the intestines of diseased laying hens in a layer farm,the inhibitory effect of Lactobacillus plantarum strain ZG-7 on laying hen-derived Clostridium perfringens was evaluated in vitro.Through co-culture experiments,the agar diffusion method,determination of acid-producing capacity,and analysis of the pH stability,enzyme treatment,and organic acid components of its cell-free supernatant(CFS),the antibacterial activity and the material basis of its action were investigated.The results showed that strain SW in the co-culture system was significantly inhibited after 4 hours of growth(P<0.05),and bacterial growth became extremely slow by 6 hours.The original CFS of strain LP ZG-7 produced a clear inhibition zone(16.4 mm)against SW.The size of the inhibition zone of CFS adjusted to pH 3 showed little change(16.5 mm),while the diameter of the inhibition zone of CFS adjusted to pH 7 decreased to 5.5 mm,indicating that the antibacterial activity was derived from acidic substances.Treatment with proteinase K almost completely eliminated the antibacterial activity of CFS.Treatment with trypsin and pepsin resulted in an inhibition zone of approximately 10 mm,while the supernatant without enzyme treatment had an inhibition zone diameter of about 16 mm.This suggests the presence of proteinaceous antibacterial substances sensitive to proteases in the CFS of Lactobacillus plantarum.Lactobacillus plantarum ZG-7 could rapidly reduce the pH of MRS medium from 6.3 to below 4.0 within 6 hours of culture,with a final pH of approximately 3.7 at 24 hours,demonstrating strong acid-producing capacity.In conclusion,Lactobacillus plantarum ZG-7 exhibits efficient inhibitory effects on Clostridium perfringens in vitro through acid production and secretion of bacteriocin-like substances,showing promising application prospects as a potential probiotic candidate strain for the prevention of necrotic enteritis.
