红豆杉内生真菌Penicillium sp.HDS-P-2次级代谢产物的分离与鉴定

Isolation and identification of secondary metabolites of the endophytic fungus Penicillium sp.HDS-P-2 from Taxus chinensis

摘要 目的研究红豆杉[Taxus chinensis(Pilg.)Rehder]内生真菌Penicillium sp.HDS-P-2中的次级代谢产物,评价化合物的肿瘤细胞毒作用。方法采用硅胶、ODS、Sephadex LH-20凝胶、MCI柱色谱及高效液相色谱等手段进行分离纯化,根据理化性质和波谱数据鉴定化合物的结构。以顺铂为阳性对照药,通过CCK-8法测定化合物对HL-60细胞的生长抑制作用。结果分离得到16个化合物,分别鉴定为penimeroterpenoid A(1)、peniandrastin G(2)、andrastin A(3)、shearinine S(4)、shearinine E(5)、shearinine A(6)、22,23-dehydroshearinine A(7)、shearinine D(8)、hypoxyterpoid A(9)、paecilomycine B(10)、penioxalicin(11)、7-羟基-4-甲氧基-5-甲基香豆素(12)、硫赭曲菌素(13)、O-methylcurvulinic acid(14)、aspernigrin A(15)、pyrophen(16)。化合物5~8对HL-60细胞的IC50为10~20μmol·L^(-1)。结论化合物12为首次从青霉属真菌中分离得到,化合物2、4、7、10、11为首次从红豆杉内生菌中分离得到。并且化合物1、5~8对HL-60细胞表现出不同程度的抑制作用。 Objective To study the secondary metabolites of the endophytic fungus Penicillium sp.HDS-P-2 from Taxus chinensis(Pilg.)Rehder,and the cytotoxic activity of the compounds against HL-60 cells.Methods The compounds were isolated by silica gel,ODS,Sephadex LH-20,MCI column chromatography and HPLC,and their structures were identified by spectral analysis.The CCK-8 assay was used to evaluate the cytotoxicity of the compounds against HL-60 cells,and cisplatin was used as a positive control.Results Sixteen compounds were isolated and identified as penimeroterpenoid A(1),peniandrastin G(2),andrastin A(3),shearinine S(4),shearinine E(5),shearinine A(6),22,23-dehydroshearinine A(7),shearinine D(8),hypoxyterpoid A(9),paecilomycine B(10),penioxalicin(11),7-hydroxy-4-methoxy-5-methylcoumarin(12),sulochrin(13),O-methylcurvulinic acid(14),aspernigrin A(15),and pyrophen(16).Compounds 5-8 displayed cytotoxic activity on HL-60 cells with IC_(50)values of 10-20μmol·L^(-1).Conclusion Compound 12 was obtained from Penicillium sp.for the first time and compounds 2,4,7,10 and 11 were obtained from Taxus chinensis endophytic fungus for the first time.Compounds 1,and 5-8 showed different degree of inhibitory activity against HL-60 cells.
