摘要
目的研究小米草化学成分及其抗光老化活性。方法采用D101大孔吸附树脂、硅胶、Sephadex LH-20及制备型HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用CCK-8、LDH试剂盒评估抗光老化活性。结果从中分离得到10个化合物,分别鉴定为(5-甲酰基-2-呋喃)甲基二氢阿魏酸酯(1)、十六烷-4-内酯13-O-β-D-吡喃葡萄糖(1→2)-β-D-吡喃葡萄糖苷(2)、芹菜素(3)、7 R,8R-4,7,9,9′-四羟基-3,3′-二甲氧基-8-O-4′-新木脂素(4)、布卢门醇A(5)、阿魏酸(6)、3-苯基乳酸(7)、3-甲氧基-4-羟基苯甲酸(8)、环(脯氨酸-丙氨酸)(9)、5-羟甲基2-呋喃甲醛(10)。化合物1~2可显著提高UVB照射下HaCaT细胞活性,抑制乳酸脱氢酶释放。结论化合物1~2为新化合物,3~10为首次从该植物中分离得到。化合物1~2具有抗光老化活性。
AIM To study the chemical constituents from Euphrasia pectinata Ten.and their anti-photoaging activity.METHODS The separation and purification were carried out by using D101 macroporous adsorption resin,silica gel,Sephadex LH-20 and preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The anti-photoaging activity was evaluated by CCK-8 method and LDH kits.RESULTS Ten compounds were isolated and identified as(5-formyl-2-furanyl)methyl dihydrof erulic acid ester(1),16 alkane-4-olide-13-O-β-D-glucopyranosy(1→2)-β-D-glucopyranoside(2),apigenin(3),(7 R,8R-4,7,9,9′-tetrahydroxy-3,3′-dimethoxy-8-O-4′-neolignan)(4),blumenol A(5),ferulic acid(6),p-coumaric acid(7),3-methoxy-4-hydroxybenzo(8),cyclo(Pro-Ala)(9),5-hydroxylmethyl-2-furfural(10).Compounds 1-2 could significantly enhance the viability of HaCaT cells under UVB irradiation,and inhibit the release of lactate dehydrogenase.CONCLUSION Compounds 1-2 are new compounds,3-10 are isolated from this plant for the first time.Compounds 1-2 have anti-photoaging activity.
