摘要
目的:研究乳香的化学成分。方法:采用硅胶、Sephadex LH-20葡聚糖凝胶、ODS柱及制备型HPLC等色谱技术进行分离纯化,结合1D/2D NMR、HR-MS等波谱数据对化合物的结构进行鉴定。采用CCK-8法筛选化合物对人肝癌HepG2细胞的抗肿瘤活性。结果:从乳香70%乙醇提取物中分离得到10个化合物,分别鉴定为:3α-乙酰氧基-11,16-二羰基-β-乳香酸(1)、9,11-去氢-β-乳香酸(2)、β-乳香酸(3)、11-羰基-β-乳香酸(4)、3α-乙酰氧基-11-羰基-β-乳香酸(5)、3α-乙酰氧基-11α-甲氧基-β-乳香酸(6)、3β-acetoxyurs-12-en-11-one(7)、3α-乙酰氧基-9,11-去氢-α-乳香酸(8)、α-乳香酸(9)、3,11-dioxo-24-norurs-12-en(10)。化合物4和6对人肝癌HepG2细胞的IC_(50)分别为(57.70±5.34)μmol/L和(40.00±14.47)μmol/L。结论:其中,化合物1为新化合物,化合物10为新天然产物;化合物4和6对人肝肿瘤细胞的增殖具有一定的抑制作用。
Objective:To study the chemical constituents of Boswellia carterii.Methods:The compounds were isolated and purified by the silica gel,Sephadex LH-20,ODS,pre-HPLC.The structures were identified by 1D/2D NMR and HR-MS spectroscopic data.The antitumor activity of the compounds against human hepatocellular carcinoma HepG2 cells was screened using the CCK-8 method.Results:Ten compounds were isolated from 70%ethanol extract of Boswellia carterii and identified as 3α-O-acetoxy-11,16-diketo-β-boswellic acid(1),9,11-dehydro-β-boswellic acid(2),β-boswellic acid(3),3α-hydroxy-11-keto-β-boswellic acid(4),3α-acetoxy-11-keto-β-boswellic acid(5),3α-O-acetoxy-11α-methoxyl-boswellic acid(6),3β-acetoxyurs-12-en-11-one(7),3α-O-acetoxy-9,11-dehydro-α-boswellic acid(8),α-boswellic acid(9),3,11-dioxo-24-norurs-12-en(10),respectively.The IC_(50) of compounds 4 and 6 against human liver cancer HepG2 cells were(57.70±5.34)μmol/L and(40.00±14.47)μmol/L.Conclusion:Among them,compound 1 is a new compound.Compound 10 is a new natural products.compounds 4 and 6 exhibit certain inhibitory effects on the proliferation of liver tumor cells.
