UPLC-Q-TOF-MS/MS法鉴定3种酚类在小鼠体内的代谢产物

摘要 目的建立UPLC-Q-TOF-MS/MS法鉴定没食子酸、鞣花酸、柯里拉京在小鼠体内的代谢产物。方法给药组小鼠分别灌胃给予3种成分的0.5%CMC-Na混悬液(400 mg/kg),空白组小鼠灌胃给予0.5%CMC-Na混悬液(400 mg/kg),采集血浆、尿液、粪便。分析采用Acquity UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8μm);流动相水(含0.1%甲酸)-乙腈,梯度洗脱;体积流量0.27 mL/min;柱温30℃;电喷雾离子源;负离子扫描。结果共鉴定出27种没食子酸代谢产物、13种鞣花酸代谢产物、37种柯里拉京代谢产物,这3种成分主要发生水解、脱羧等Ⅰ相代谢,以及甲基化、硫酸化、葡萄糖醛酸化等Ⅱ相代谢。结论该方法稳定可靠,可为探究其他天然酚类的体内代谢规律提供参考。 AIM To establish a UPLC-Q-TOF-MS/MS method for the identification of in vivo metabolites of gallic acid,ellagic acid and corilagin in mice.METHODS The mice in administration groups were given intragastric administration of 0.5%CMC-Na suspensions of 3 constituents(400 mg/kg),respectively,while those in blank group were given intragastric administration of 0.5%CMC-Na suspensions(400 mg/kg),after which plasma,urine and feces were collected.The analysis was performed on a 30℃ thermostatic Acquity UPLC HSS T3 column(100 mm×2.1 mm,1.8μm),with the mobile phase comprising of water(containing 0.1%formic acid)-acetonitrile flowing at 0.27 mL/min in a gradient elution manner,and electrospray ionization source was adopted in negative ion scanning.RESULTS Total 27 gallic acid metabolites,13 ellagic acid metabolites and 37 corilagin metabolites were identified,these 3 constituents mainly occured phaseⅠmetabolites such as hydrolysis,decarboxylation,and phaseⅡmetabolites such as methylation,sulfation,glucuronidation.CONCLUSION This stable and reliabe method can provide a reference for exploring the in vivo metabolic rules of other natural phenols.
